摘 要:转红色荧光蛋白基因唐鱼(Tanichthys albonubes)测交获得实验鱼,以表达红色荧光蛋白为表型性状,采用点杂交和遗传学分析相结合的方法,研究外源基因的遗传传递规律,并对转基因唐鱼和非转基因唐鱼的生长和繁殖等方面进行了比较。结果显示:测交后代表型的分离规律基本符合经典的孟德尔单显性基因遗传模式,外源基因属于单位点整合,测交后代中没有发现沉默整合个体;实验鱼的生长和可量性状对比实验表明,转基因唐鱼和非转基因唐鱼在仔鱼期到性成熟之间,生长速度无显著差异,性成熟个体的外部形态除体色外也无差别,繁殖力方面基本无显著差异。本研究结果表明了培育转红色荧光蛋白基因唐鱼纯系的可行性;同时在生长、繁殖方面的研究结果为评价转基因唐鱼的生态安全奠定了基础。Based on successfully gained transgenic Tanichthys albonubes expressing red fluorescent proteins, we carried out genetic analysis combined with dot blot to investigate the genetic transmission rule of exogenous gene in transgenic fish. We also compared growth and reproduction between transgenic Tanichthys albonubes and non-transgenic fish. The results showed that the transgenic fish inherited its unique color directly from their parents, and passed the color along to their offspring whose phenotype according to Mendel's law of segregation. The exogenous fluorescent gene was integrated in single insertion, and there were no individuals which had the integration of silence in test crossing progenies. The growth and quantitative character contrast test from one family indicated that there were no significant differences in growth between non-transgenic Tanichthys albonubes and transgenic fish from juvenile to sexual maturity, in addition, there were also no differences in external morphology between sexually mature individuals from morphological characters contrast test. The contrast test on fecundity indicated that there were no significant differences among four groups which included fecundity and hatchability, while the rate of fertilization in group D ( Tg fish × Tg fish) was lower than group A ( non-Tg fish×non-Tg fish) and group C ( Tg fish× non-Tg fish). Results of the study indicate as follows: it is possible to culture the pure line transgenic Tanichthys albonubes expressing red fluorescent proteins; transgenic fish do not have any apparent competitiveness advantage over non-transgenic fish on growth and reproduction, which provide foundation for evaluation of ecological impact of transgenic Tanichthys albonubes when they are accidentally released to the wild
