摘 要:为了解瓯江彩鲤红、白两种体色差异的分子基础,应用抑制性差减杂交(SSH)技术对这两种体色的差异表达基因进行了初步研究,以"全红"体色(全身体表为红色)彩鲤的cDNA为检验cDNA(Tester),以"粉玉"体色(全身体表为粉白色)彩鲤的cDNA为比对cDNA(Driver),进行抑制差减杂交。对筛选出的42个阳性克隆进行测序和序列比对,发现16条EST序列,其余26个克隆为空载体序列。在16条EST序列中,6条为鲤的线粒体DNA序列(鲤的线粒体:16S rRNA序列,COⅢ),5条为斑马鱼的相关基因序列(斑马鱼的骺蛋白聚糖、核心蛋白聚糖、光蛋白聚糖等相关基因;斑马鱼新型免疫型受体基因,Olfm1蛋白合成基因;斑马鱼原骨胶原蛋白mRNA),1条为鲫的mRNA序列,2条与无脊椎动物基因相类似(利氏曼原虫的22号基因组基因;蝇的抗菌缩氨酸前体mRNA基因),另2条为未知基因序列;比对结果发现4条序列可能与体色有关。为进一步筛选鲤体色调控相关的功能基因奠定了基础。In order to understand the molecular basis of the red and white color patterns in Oujiang color common carp (C. carpio var. color), a suppressive subtractive hybridization(SSH) method was used to study the differentially expressed genes of body color patterns of this species in this study. The cDNA of "Quanhong" body color (whole red of body surface) was used as the Tester, and the cDNA of " Fenyu" body color (whole white of body surface) was used as the Driver. The total of 42 clones were sequenced, in which 16 clones were the ESTs and 26 clones vector sequences. In the 16 ESTs, there were 6 mitochondrial genes of common carp (Cyprinus carpio complete mitochondrial genome: 16S ribosomal RNA gene, COU[ ) , 5 protein genes of Zebrafish ( Zebrafish DNA sequence : the epyc gene for epiphycan, the dcn gene for decorin,the gene for a novel protein similar to vertebrate Lumican; Zebrafish novel immune-type receptor (NITR) genes, Olfactomedin 1 gene ; Danio rerio mRNA for procollagen type I alpha 2 chain) , 1 mRNA sequence of crucian carp, 2 invertebrate genes (eg. Leishmania infantum and Drosophila ), and 2 unknown new genes. Four sequences may be associated with the body color of the color common carp. The study is useful for further studying the relationship of functional genes and body color patterns in Oujiang color common carp.
