摘 要:依据梨S-RNase基因保守区设计引物,对23个梨品种(类型)基因组进行了PCR扩增、PCR扩增产物克隆、测序和序列比对分析。根据S基因核苷酸序列同源性100%原理,鉴定出20个S等位基因,其中17个S基因与GenBank中已知的Sb,Sc,Sd,Sh,S8,S12,S13,S16,S19,S27,S29,S30,S31,S34,S36,S41,S42-RNases相同;在小香水中鉴定出1个新的S-RNase基因,片段大小为996bp,现已提交Genbank,登录号为FJ490411,暂未定名;寒红梨和早梨18中各有1条S基因没有被鉴别出。在此基础上,确定了20个秋子梨品种(类型)S基因型,分别是:谢花甜(S29S34),山鸭梨(S30S36),苹香梨(SdS31),马蹄黄(S16S19),寒香梨(S12S31),龙香(S12S16),早白(S19S42),八里香(S19S30),尖把子(S27Sh),软把子(S16S36),糖梨(S27S30),酸梨锅子(S19S41),延边大香水(SdS12),南果梨(S13S34),大南果(S13S34),油红(S13S34),小香水(S27Sx),野生类型山梨1(S8S27),山梨3(SbS41),山梨4(ScS42);1个白梨品种大慈梨(S19S27);秋子梨品种寒红梨和沙梨品种早梨18各鉴定出1条S基因,S基因型分别是S27Sx和SdSx。Total of 23 cultivars and wild forms of Pyrus ussuriensis Maxim.were used as material for identifying their S-geno-type using peculiar primers designed by the conserved regions of pear S-RNase structure,PCR amplifications,amplification product cloning,sequencing and sequences alignment analysis.Based on principle of homology of 100% of S gene nucleotide sequence,20 S-RNase genes were determined;17 S-RNase genes of them,Sb,Sc,Sd,Sh,S8,S12,S13,S16,S19,S27,S29,S30,S31,S34,S36,S41,S42,were the same as these known on the GenBank;one novel S-RNase was discovered in pear cultivar Xiaoxiangshui,PCR bands 509 bp,accession numbers FJ490411;One of S gene from Hanhongli and Zaoli 18 was unidentified respectively.The S-genotypes of 20 pear cultivars were identified as follows:Xiehuatian(S29S34),Shanyali(S30S36),Pingxiangli(SdS31),Matihuang(S16S19),Hanxiang(S12S31),Longxiang(S12S16),Zaoxiang(S19S42),Balixiang(S19S30),Jian-bazi(S27Sh),Ruanbazi(S16S36),Tangli(S27S30),Suanliguozi(S19S41),Yanbian Daxiangshui(SdS12),Nanguoli(S13S34),Dananguo(S13S34),Youhong(S13S34),Xiaoxiangshui(S27Sx),wild type Shanli 1(S8S27),wild type Shanli 3(SbS41),wild type Shanli 4(ScS42).The S-genotypes of Dacili(P.bretschneideri) was S19S27,Hanhongli(P.ussuriensis) was S27Sx,Zaoli 18(P.pyrifolia) was SdSx.
