摘  要:目的探讨黄芪多糖(APS)对糖尿病心肌病变干预机制。方法检测APS治疗组和糖尿病(DM)对照组仓鼠糖化血清蛋白、胰岛素、C肽、心肌酶谱水平,电镜观察心肌组织超微结构,免疫组化检测心肌Ⅰ、Ⅲ型胶原,放射免疫法测定血浆和心肌血管紧张素Ⅱ(ATⅡ)含量,荧光定量RT-PCR法检测心肌胃促胰酶和血管紧张素转化酶(ACE)表达,放射免疫法检测胃促胰酶和ACE活性,WesternBlot法检测心肌磷酸化的细胞外信号调节激酶1/2(p-ERK1/2)含量。结果与DM组相比,APS组糖化血清蛋白、心肌酶谱、心肌ATⅡ及心肌胶原含量均下降,心肌病变减轻,心肌胃促胰酶表达和活性均下降,心肌p-ERK1/2含量降低,ACE基因表达和活性无统计学差异。结论 APS可抑制糖尿病仓鼠心肌胃促胰酶依赖性ATⅡ生成,从而保护心肌。Objective To investigate effects of astragalus polysaccharides(APS)on diabetic hamster cardiomyopathy. Methods Insulin, C-peptide, myocardial enzymes, glycosylated serum protein(GSP), AT Ⅱ in plasma and myocardium were tested. Myocardial uhrastructure and collagen were studied. Expressions of chymase and ACE mRNA were detected by real-time fluorescent quantitative RT-PCR, and their protein activities were tested. Expressions of phosphoric extracellular signal-regulated kinase 1/2 (p- ERK1/2)in hearts were measured by Western blot in diabetic hamsters administrated with APS or normal saline(NS). Results Levels of GSP, CK, CK-MB and myocardial AT Ⅱ in APS-treated group were lower than those in control group. Positive index of myocardial collagen Ⅰ in APS treatment group was less than that in control group. Myocardial ultrastructure was well-protected in APS treatment group compared with control group. Myocardial chymase expression and activity,p-ERK1/2 expression in APS treatment group were lower than those in control group. While levels of insulin, C-peptide, plasma AT Ⅱ and cardial ACE expression and activity were similar in two groups. Conclusions APS is effective in treating diabetic cardiomyopathy due to inhibition of local chymase activity.
