耐环丙沙星沙门菌marA、soxS和ramA基因以及外排泵基因acrA和acrB的表达水平分析

摘　　要：应用实时荧光定量PCR法对11株鸡源环丙沙星诱导耐药沙门菌和11株人源临床环丙沙星耐药沙门菌的调控基因marAs、oxSr、amA和外排泵基因acrA、acrB的mRNA表达水平进行了检测和分析。结果表明,11株诱导株与诱导母株相比,marA、soxS、acrA和acrB的mRNA表达量均显著增加(P<0.05),ramA表达量未增加;11株人源临床耐药株与标准菌株相比,marA、acrA和acrB的表达量显著增加(P<0.05),soxS和ramA的表达量未增加。提示,鸡源诱导株AcrAB过表达与marA和soxS过表达相关;人源临床株AcrAB过表达与marA过表达相关,而与soxS和ramA表达不相关。The expression levels of regulatory genes marA ,soxS and ramA and efflux pump genes acrA and acrB were determined by real-time fluorescent quantitative PCR in 11 in vitro selected chicken-derived ciprofloxacin resistant mutants and 11 human derived clinical isolates. Significant increases in mRNA expression levels of marA,soxS,acrA and acrB were observed in the 11 mutants compared with their parent strains（P〈0.05） ,and there was no significantly difference with regard to the expression of ramA. The expression of marA,acrA and acrB in the 11 human derived clinical isolates also increased significantly（P〈0.05） when compared with the reference strain. However, there was no significantly difference in the expression of soxS and ramA. The results indicated that AcrAB over expression in the mutants might correlate with marA and soxS over-expressions. AcrAB ove〉expression in human-derived clinical isolates might correlate with marA over-expression and not be related to soxS and ramA over-expressions.
