补气固表制剂对小鼠CD_4^+CD_(25)^+Treg的增殖作用体外实验研究

摘　　要：目的观察补气固表中药制剂对小鼠胸腺CD4+CD25+Tregs增殖活性的促进作用。方法取小鼠胸腺,分离种植细胞,体外培养,以不同浓度的补气固表中药制剂进行处理后,进行CD4+CD25+Treg抗体双标记,流式细胞仪检测Tregs增殖活性,比较不同组别的增值活性差异。结果检测结果显示,应用不同浓度补气固表中药制剂干预处理后,对CD4+CD25+Treg增值活性均有明显的促进作用(P<0.01),尤以中浓度组的效应更为显著(P<0.01)。结论体外实验证实,补气固表中药制剂对小鼠胸腺CD4+CD25+Tregs的增殖活性有刺激作用,为从调控Tregs增值活性角度治疗变态反应性疾病提供了实验依据。Objective To investigate the promoting effect of Qi-boosting Exterior-securing herbal medicine preparation on the proliferating activity of CD4^＋CD25^＋Tregs of mice based on an experimental study in vitro. Methods Thymus of was taken from mice at first, T cells were separated from thymus, and then, cells were cultivated in vitro with Qi-boosting Exterior-securing herbal medicine preparation at different concentration levels for several hours. Followed were the procedures of CD4^＋CD25^＋Tregs stained by double antibody labeling and detected by flow cytometry to evaluate the differences in the proliferating activities among different groups of CD4^＋CD25^＋Tregs treated with Qi-boosting Exterior-securing herbal medicine preparation at different concentrations. Results It was shown that Qi-boosting Exterior-securing herbal medicine preparation could significantly promote the proliferating activities of CD4^＋CD25^＋Tregs at all the concentrations tested in this study （P〈0.01）, but with the most significant effect as determined in the group cultured with the moderate concentration of herbal medicine preparation （P〈0.01）, when compared with that of controlling group. Conclusions As determined in this experimental study, Qi-boosting Exterior-securing herbal medicine preparation can effectively activate CD4^＋CD25^＋Tregs in vitro separated from the thymus of mice. These results may provide useful experimental evidences for the feasibility to explore therapy for the treatment of allergic diseases through regulating the proliferating activity of Tregs by reagents such as Qi-boosting Exterior-securing herbal medicine preparation.
