摘  要:目的探讨体外分离培养的人尿道黏膜上皮细胞的生物学特性,为构建组织工程尿道提供种子细胞。方法取尿道下裂患者尿道黏膜,经消化离心后,接种于培养基中连续培养,观察细胞形态变化及生长、增殖过程,并绘制生长曲线;对细胞进行常规冻存和复苏;应用免疫细胞化学技术进行角蛋白染色的鉴定。结果体外培养的细胞呈铺路石样外观,可传至8代;可进行常规冻存和复苏;角蛋白免疫细胞化学染色呈阳性。结论人尿道黏膜上皮细胞的体外连续培养及扩增技术被建立,本实验培养的细胞可用于构建组织工程尿道。Objective To investigate the biologic characteristics of human urethral epithelium cuhured in vitro, and to provide seed cells for tissue engineering urethra. Methods Specimens from urthral epithelium of hypospadias were isolated, digested and cultured. The cell morphology, growth and proliferation were observed. Also, the urethral epithelial cells were cryopreserved stored for further study. Besides, the cells were stained by immunocytochemistry to detect the expression of cytokeratin. Results The cultured urethral epithelium cells could be subcuhured to 8 generations, they were the same in size like road rocks. The preservation and recovery of cells were achieved. The cells showed positive staining for cytokeratin. Conclution The culture in series and expansion technique for human urethral epithelium has been established. Cells cultured in this experiment could be used in construction of tissue engineering urethra.
