2,5-二羟苯磺酸钙对糖尿病大鼠心肌保护作用的研究

摘　　要：目的观察2,5-二羟苯磺酸钙对链脲佐菌素(streptozotocin,STZ)诱导的糖尿病大鼠心肌病变的保护作用。方法将24只Wistar大鼠随机分为正常对照组、糖尿病模型组和羟苯磺酸钙治疗组,每组8只。给药8周后处死大鼠,观察心肌组织病理变化和超微结构,并用免疫组化法测定心肌纤溶酶原激活物抑制物-1(PAI-1)和基质金属蛋白酶-9(MMP-9)的表达。结果糖尿病大鼠心肌纤维排列紊乱,线粒体肿胀、聚集。VG染色显示,糖尿病大鼠心肌间质胶原纤维明显增多,免疫组化示PAI-1表达明显增多,MMP-9降低。羟苯磺酸钙治疗组大鼠上述趋势均明显好转。结论羟苯磺酸钙可能通过抑制糖尿病大鼠心肌间质纤维化,延缓糖尿病心肌病变的发展。People＇s Hospital,Wendeng 264422,Shandong,China） Objective To explore the effect of calcium dobesilate on diabetic rats induced by streptozotocin.Methods Wistar rats were randomly divided into the normal control group（N）,the diabetic group（DM）and the diabetic group treated with calcium dobesilate（T）.Rats in the T group were fed with calcium dobesilate for 8 weeks.At the end of the 8th week,rats were killed for light microscopy and electron microscopy.Then expressions of PAI-1 and MMP-9 were determined by immunohistochemistry.Results Compared with the N group,rats in the DM group had abnormal cardiac ultrastructure with confusion of myofibril and swelling and gathering of mitochondria.VG staining showed that the collagen content was increased in diabetic rat hearts.Expression of PAI-1 was increased in diabetic myocardium,while expression of MMP-9 was obviously decreased.The above trends were significantly improved in the T group.Conclusion Calcium dobesilate postpones the development of diabetic cardiomyopathy through inhibiting myocardial fibrosis.
