摘 要:目的:考察川芎嗪对辐射致血虚证小鼠骨髓细胞IL-4R和IL-7R基因表达的影响,为阐明川芎嗪的补血机制提供依据。方法:采用3.5Gy^(60)Coγ射线全身1次性照射制备小鼠血虚证模型;骨髓集落培养计数CFU-GM、CFU-E、BFU-E、CFU-mix;RT-PCR检测骨髓IL-4R和IL-7R基因表达。结果:川芎嗪显著升高放射线致血虚证小鼠骨髓CFU-GM、CFU-E、BFU-E、CFU-mix数量,上调骨髓细胞IL-4R和IL-7R基因表达。结论:川芎嗪促进骨髓细胞IL-4R和IL-7R基因表达可能是其升血和调节免疫功能的机制之一。Objective:To observe the effect of ligustrazine on IL-4R and IL-7R gene expression in bone marrow cell of blood deficiency mice induced by radiation and to provide the foandation for explaining the blood enriching mechanism of ligustrazine.Methods:The blood deficiency mice were induced by using a single dose of 3.5 Gy 60Co γ radiation.Myeloid colony cell was cultured and the number of CFU- GM,CFU-E,BFU-E and CFU-mix were counted.IL-4R and IL-7R gene expression in bone marrow were measured by RT-PCR.Results: Ligustrazine can raise significantly the number of CFU-GM,CFU-E,BFU-E and CFU-mix in bone marrow of blood deficiency mice and upregulate IL-4R and IL-TR gene expression of bone marrow cett.Conclusions:Ligustrazine can promote IL-4R and IL-7R gene expression in bone marrow cells,which probably is one of the mechanisms of blood proliferation and immune regulation.
