豆野螟颗粒体病毒的分离鉴定

摘　　要：作者采用差异离心术、超薄切片与电镜术对豆野螟颗粒体病毒进行分离纯化、组织病理及超微结构研究。发现颗粒体多呈椭圆形，直径大小３００～５００ｎｍ×２００～３００ｎｍ；病毒粒子呈杆状，稍弯曲，大小２００～３００ｎｍ×４０～５０ｎｍ。该病毒侵染宿主脂肪体，表皮、中肠上皮、气管、马氏管等组织的细胞核，并观察到颗粒体病毒在细胞核中复制的过程。The bean pod borer, Maruca testulalis Greyer granulosis virus (MtGV) was purified with differential centrifugation on sucrose gradients. Electron microscopical studies on the histopathology and ultrastructure of MtGV showed that the majority of the capsules measured about 300~500 nm×200~300 nm and were mostly ovocylindrical in shape. The dimensions of the virus rods of MtGV were 200~300 nm in length by 40~50 nm in diameter. In addition to the fat body, MtGV infected the hypodermis, midgut epithelium, tracheal matrix, and malpighian tubules of the host insect larvae. The replication cycle of MtGV in the cell nucleus was also observed.
