摘 要:目的:观察西洋参治疗胰岛素抵抗(Insulin Resistance,IR)大鼠活性部位对Resistin基因表达的影响。方法:通过早期药效学试验证明,西洋参根部60%洗脱部分和30%洗脱部分为有效部位。本实验采用高脂饲料喂养加链脲佐菌素法复制IR大鼠(SD)的动物模型,造模成功后,将大鼠随机分为模型组、罗格列酮组、西洋参60%洗脱组、西洋参30%洗脱组。给药4周后,以RT-PCR法检测附睾周围白色脂肪组织中Resistin-mRNA的表达。结果:与模型组比较,各给药组大鼠脂肪组织中Resistin基因mRNA的表达均降低,其中以西洋参60%洗脱组作用最强,与模型组比较有极显著差异(P<0.01),西洋参30%洗脱组和罗格列酮组与模型组比较差异显著(P<0.05)。结论:西洋参可降低IR大鼠脂肪组织中Resistin基因的表达;西洋参60%洗脱部位降低Resistin基因表达的作用最明显。Objective:This study focuses on the impact of active fraction of American Ginseng on the expression of treating insulin resistance rat 's Resistin -mRNA. Methods : It was proved before that 30% alcoholic extraction of American Ginseng and 60% alcoholic extraction of Ameri-can Ginseng are effeetive component by pharmacodynamic studies. In this study, SD rats are raised by high tat diet combined with STZ in or-der to produce insulin resistance rat model. Insulin resistance rats are divided randomly into model group, rogridone group, 30% alcoholic extraction of American Ginseng group and 60% alcoholic extraction of American Ginseng group. Four weeks later, these groups are detected the expression of Resistin -mRNA by RT -PCR. Results: Compared with model group, the expression other group rats' Resistin -mRNA is decreased. There was an extreme difference between Ginseng 60% al-coholic extraction group and model group( P <0.01 ). There was a sig-nificant difference between Ginseng 30% alcoholic extraction group and model group( P < 0.05 ). Conclusion : Insulin resistance rats' Resistin -mRNA expression can be decreased by American Ginseng , 60% al-coholic extraction of American Ginseng group has a bigger impact on the expression of Resistin -mRNA.
