子痫前期患者胎盘组织中促炎与抗炎细胞因子mRNA的表达及意义

摘　　要：目的探讨促炎细胞因子与抗炎细胞因子mRNA在子痫前期患者胎盘组织中的表达及意义。方法采用RT-PCR技术测定36例正常足月妊娠妇女(A组)和24例轻度子痫前期患者(B组)及17例重度子痫前期患者(C组)胎盘组织中肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1、IL-4、IL-6、IL-10 mRNA的表达水平。结果C组TNF-α、IL-6、IL-1 mRNA表达水平分别为(1.81±0.23)、(1.31±0.19)和(0.99±0.16),显著高于B组的(0.98±0.18)、(1.07±0.15)和(0.61±0.12)及A组的(0.92±0.14)、(0.72±0.11)和(0.65±0.11)(P<0.05)。C组IL-4、IL-10 mRNA表达水平分别为(0.38±0.09)和(0.25±0.15),显著低于B组的(0.59±0.16)和(0.83±0.20)及A组的(0.81±0.13)和(0.94±0.25)(P<0.05)。B组IL-6 mRNA表达水平显著高于A组(P<0.05);B组IL-4 mRNA表达水平显著低于A组(P<0.05)。结论子痫前期患者胎盘组织中促炎细胞因子TNF-α、IL-1、IL-6 mRNA表达水平上调,抗炎细胞因子IL-4、IL-10 mRNA表达水平降低。Objective To study the expression and significance of pro- and anti-inflammatory cytokines in placenta in preeclampsia（PE） patients. Methods The levels of tumor necrosis factor （TNF）-α, interleutin（IL）-1, IL-4, IL-6 and IL-10 mRNA were measured by semi-quantitative RT-PCR in the placenta of 36 normal pregnancy woman（group A）,24 cases of mild PE（group B） and 17 cases of severe PE（group C）. Results The levels of TNF-α, IL-1 and IL-6 mRNA were （1.81±0.23）, （1.31±0. 19） and （0.99±0.16） ,respectively,in group C,which were significantly higher than （0. 98 ±0. 18）, （1.07±0. 15） and （0.61±0. 12） in group B and （0. 92±0.14）, （0. 72±0. 11） and （0. 65± 0. 11） in group C （P〈0. 05）. The levels of IL-4 and IL-10 mRNA were （0. 38±0.09） and （0. 25± 0. 15） in group C, which were significantly lower than （0. 59±0. 16） and （0. 83±0. 20） in group B and （0. 81±0.13） and （0.94±0. 25） in group A （P〈0. 05）. The level of IL-6 mRNA in group B was significantly higher than that in group A （P〈 0. 05 ）. The level of IL-4 m.RNA in group B was significantly lower than that in group A （P〈0. 05）. Conclusion The expression of pro-inflammatory cytokines of TNF-α, IL-1 and IL-6 are up-regulated in PE placenta and anti-inflammatory cytokines of IL-4 and IL-10 are down-regulated.
