摘 要:以质粒pBR322为对象,在经典的质粒提取方法的基础上进行了一些改进,将酚氯仿异戊醇抽提和氯仿异戊醇抽提省去。比较了简便方法和经典方法提取质粒的效果,结果显示:两种方法均能够提取出质粒DNA,RNA的量并没有比经典方法提出来的多;简便方法抽取的质粒,蛋白含量稍稍多于后者;简便方法节省了大约一半的提取时间及大量的药品,并且减少了有害的化学物质对人体的危害,是一种经济实用的方法,完全可以满足一般实验需要。Plasmid pBR322 was used in this study and some improvement wew made based on the classical extraction method. The step of extracting by phenol chloroform isoamyl alcohol and chloroform isoamyl alcohol was omitted. Comparing the result of this two extraction method, it can find that the two methods both can get plasmid DNA and the quantity of RNA was no more than it by using the classical way. After omitting the two extraction method, the amount of protein is a little more than classical method. On one hand it will save about a half time, on the other hand it also can save plenty of physic. It will decrease the harm to human. This is an economical and convenient method and it can absolutely match the need of general experiment.
