摘 要:目的:研究姜黄素固体分散体(SDs,姜黄素:聚乙烯吡咯烷酮PVP k30质量比1∶8)对大鼠胃溃疡的作用。方法:大鼠设为模型组、姜黄素SDs低、中、高剂量(含姜黄素10,30,90 mg.kg-1)组,阳性对照雷尼替丁(27 mg.kg-1)组。每组10只。乙酸致大鼠胃溃疡模型,连续口服给药14 d后,取血测定血清NO、血浆ET,测定胃溃疡指数;制备幽门结扎致大鼠胃溃疡模型,给药3 d后禁食36 h麻醉条件下施幽门结扎术,16 h后解剖收集胃液,测定胃液量、胃蛋白酶活性、溃疡指数;小鼠口服给药3 d后禁食24 h,第5天皮下注射利血平10 mg.kg-1制备胃溃疡模型,6 h后处死测定胃溃疡指数。结果:乙酸致胃溃疡实验表明,与模型组溃疡指数5.87±0.48,血清NO(23.63±5.73)mmol.L-1相比,口服姜黄素SDs中、高剂量组大鼠溃疡指数显著降低,分别为4.59±0.96,3.33±0.93(P<0.01),大鼠血清NO水平明显升高,分别为(29.75±5.90)mmol.L-1(P<0.05),(39.63±12.73)mmol.L-1(P<0.01);与模型组血浆ET(163.65±63.84)ng.L-1相比,姜黄素SDs高剂量明显降低大鼠的血浆ET水平(104.22±63.84)ng.L-1(P<0.05)。幽门结扎胃溃疡实验表明:与模型组溃疡指数相比,姜黄素SDs高剂量能够显著降低大鼠溃疡指数(P<0.01);与模型组胃液量、胃酸分泌以及胃蛋白酶活性相比,姜黄素SDs中、高剂量能够明显降低大鼠胃液量,分别为(12.68±1.46)mL(P<0.05),(9.99±0.79)mL(P<0.01),能够明显降低大鼠胃酸分泌(77.62±8.34)mmol.L-1(P<0.05),(65.77±8.19)mmol.L-1(P<0.01),能够明显抑制大鼠胃蛋白酶活性(358.13±37.44)U.mL-1(P<0.05),(292.13±41.93)U.mL-1(P<0.01)。利血平诱导胃溃疡模型实验表明:与模型组溃疡指数相比,姜黄素SDs中、高剂量组能够显著降低利血平诱导胃溃疡的溃疡指数,分别为3.88±0.40,3.03±0.64(P<0.01)。结论:通过多种实验性胃溃疡模型证明:姜黄素SDs具有明显的抗胃溃疡作用,通过抑制胃酸的分泌、降低胃液的酸度、抑制胃蛋白酶活性,促Objective: To research the pharmacological action of curcumin solid dispersions (SDs, curcumin and polyvinylpyrrolidone (PVP) k30 in the ratio of 1: 8) was investigated on experimental gastric ulcer in rats and mice. Method: Animals were randomly divided into several experimental groups. Each group consisted of 10 animals. The control group received PVP vehicle (720 mg·kg^-1 , po) throughout the course of the experiments. The treatment groups received different doses of curcumin SDs (equivalent to curcumin 10, 30 and 90 mg ·kg^-1 , po), and ranitidine (27 mg ·kg^-1 , po) was used as the positive control. In acetic acid-induced gastric ulcers model, serum NO, plasma ET and gastric ulcer indexes of rats were measured after oral administration for 14 d. In rat ulcer model induced by pylorus-ligature, gastric volume pepsin and gastric ulcer indexes of rats were measured after oral administration for 3 d and pyloms-ligature inducement for 16 h. Gastric ulcer indexes of mice were measurement after oral administration for 3 d and subcutaneous injection reserpine 10 mg ·kg^-1. Result: The results showed that curcumin SDs (equivalent to curcumin 30, and 90 mg ·kg^-1 , po) could reduce the ulcer indexes 4. 59 ± 0. 96 and 3. 33 ± 0. 93 (P 〈 0. 01 ), and increase serum NO level (29. 7± 5. 90) mmol· L^-1 (p 〈 0. 05) and (39. 63 ±12. 73) mmol · L^-1 (P 〈0. 01 ), compared to gastric index 5. 87 ±0. 48 and NO level (23.63 ±5.73) mmol· L^-1 in control group. Compared to plasma ET (163. 65 ±63.84) ng · L^-1 in control group, curcumin SDs (equivalent to 90 mg· kg^-1 , po) could decrease plasma ET level ( 104. 22 ±63. 84) ng· L-1 (P 〈0. 05).
