双黄补和转化生长因子β_1对牙周膜细胞的增殖分化的调控作用的比较

摘　　要：目的:探讨传统中药双黄补(黄连,黄岑,骨碎补)和转化生长因子β1对体外培养的人牙周膜细胞增殖和分化功能的影响。方法:本研究通过体外培养人牙周膜细胞(periodontal ligament cell,PDLCs),应用双黄补(黄连,黄芩,骨碎补)提取液作为辅助因子及转化生长因子β1作为生长因子,分成四组(A)双黄补(B)转化生长因子β(1C)双黄补加转化生长因子β(1D)对照组,采用噻唑蓝(MTT)比色法测定细胞增殖情况,羟脯氨酸法测定胶原蛋白占总蛋白比值。结果:双黄补组与对照组及其他各组比较,明显促进人牙周膜细胞增殖,增加胶原蛋白的比值,有显著性差异(P<0.05)。A,B,C各组组间比较无显著性差异。随着作用时间的延长,细胞的OD值逐渐增大,在第5天达到最大。结论:双黄补水提液对比转化生长因子β1有明显促进人牙周膜细胞增殖,明显提高胶原蛋白在总蛋白中的比值的作用,可作为人牙周膜细胞增殖的理想的辅助因子。Objective To observe the effects of shuanghuangbu （goldthread, skullcap, rhizome drynariae） and TGF-β1 on the proliferation and differentiation of human periodontal ligament cells （ PDLC）. Methods PDL cells were cultured in different group with different ingredient, and cell numbers were counted use MTT method and the ratio of collagen content in total protein was evaluated use hydroxyproline. Results Comparing with the control group,all groups of different ingredient stimulated the proliferation and differentiation of PDL cells and increased the ratio of collagen content in total protein. The effect of every group each have no significantly difference. Conclusion These findings suggest that, shuanghuangbu and TGF-β1 can significantly stimulated the proliferation and differentiation of PDL cells, and may play an important role in the regeneration of PDL.
