摘 要:目的:研究仙灵骨葆胶囊对去卵巢诱导的大鼠绝经后骨质疏松模型骨组织形态计量学参数的影响,揭示补肾类中药仙灵骨葆胶囊治疗骨质疏松的机制。方法:将健康雌性SD大鼠(230~280g/只)48只,随机分成4组:假手术组(sham)、模型组(OVX)、中药组(OVX+T)、西药组(OVX+E),各12只。除对照组外对其它组大鼠进行摘除卵巢手术制造绝经后骨质疏松大鼠模型。术后3个月,西药组肌注鲑降钙素针,3IU/次/只/d,其他时间灌服等量生理盐水;中药组用仙灵骨葆胶囊溶液灌胃,灌服药量每只0.5g/d;假手术组和模型组每日同时灌服等量生理盐水。介入治疗3个月后,处死所有大鼠,取出左胫骨上端骨骺松质骨部分,切片、染色,进行骨计量学静态参数和动态参数测量和计算并行各组间统计学对照。结果:造模组与空白组相比,%Tb.Ar、Tb.N减少,Tb.Sp增加,%L.Pm、BER/BS、BFR/BV、BFR/TV减小(P〈0.01);仙灵骨葆组、鲑降钙素组分别与造模组相比,%TB.Ar、Tb.N均增加,Tb.Sp均减小,%L.Pm、BFR/BS、BFR/BV、BFR/TV均增加(P〈0.01);%OC.Pm、%OC.Sur均减小(P〉0.05);仙灵骨葆组与鲑降钙组比较,静态和动态各项参数值比较接近,差异也无统计学意义。结论:补肾中药仙灵骨葆可以明显改善骨质疏松大鼠骨形态计量学指标:能够增加去势大鼠骨形成、降低骨吸收,可能是通过刺激成骨细胞生长,抑制破骨细胞活性,并抑制高骨转趋势来实现的,从而对绝经后骨质疏松有治疗作用。Objective:To explore the effect of Xianling Gubao capsules(XGC) on bone histomorphology in ovariectomized rats of osteoporosis.Methods:Forty-eight female SD rats were randomized into sham group,model group,XGC group,and western medicine group,12 rats in each group.All rats were ovariectomized to establish the osteoporosis models except rats in sham group.The osteoporosis models were successfully set up 3 months after operation.The rats in western medicine group were treated with intramuscular injection of Salcatonin (3 IU/d). The rats in XGC group received oral adminis tration of XGC (0.5 g/d), while the other rats were administrated with sodium chloride. After 3 months treatment, all the rats were sacrificed and cancellated bone of the left proximal tibias was collected for dynamic and static bone morphometry. Results: Compared to sham group, %Tb. Ar, Tb. N. %L. Pm, BER/BS, BFR/BV, and BFR/TV significantly decreased, and Tb. Sp markedly increased in model group (P〈0.01). Compared to model group, %Tb. Ar, Tb. N, %L. Pro, BFR/ BS, BFR/BV, BFR/TV obviously increased, and Tb. Sp remarkably decreased (P〈0.01) in XGC group and western medicine group. There was no significant difference between the XGC group and western medicine group. Conclusion: XGC can effectively improve the histomorphometry profile. The mechanism might be involved with stimulating osteogenesis and suppressing the cytoactive of osteoclast, thus exerts a therapeutic effect on postmenopausal osteoporosis.
