退变颈椎间盘中的IL-1β、TNFα含量变化及软骨终板钙化的动物实验研究  被引量:7

摘  要:目的:观察退变颈椎间盘内细胞因子的含量变化情况和颈椎软骨终板钙化与颈椎间盘退变的关系,研究中西药治疗退变颈椎间盘对细胞因子含量及软骨终板钙化的影响。方法:选用25只4月龄新西兰兔,建立家兔颈椎动力平衡失调模型,诱导颈椎间盘退变(正常对照组不作处理)。术后7个月,药物治疗组(浅层、全层)给予抗骨增生胶囊和葡立胶囊(剂量按体重折算),灌胃2次/d,连用1个月。1个月后用耳缘静脉气栓法处死各组动物,用双抗夹心ELISA法检测颈椎间盘中IL-1β、TNFα抗体含量,并在形态学上评定颈椎间盘退变程度,测定各组动物颈椎软骨终板钙化层厚度,进行组间比较。结果:模型浅层组、模型全层组与正常对照组相比较,颈椎间盘中IL-1β、TNFα含量明显升高(P<0.01),颈椎软骨终板钙化层明显增厚(P<0.01)。而抗骨增生胶囊与葡立胶囊联合应用治疗退变颈椎间盘对IL-1β、TNFα含量及软骨终板钙化有明显抑制作用(P<0.05)。结论:退变颈椎间盘组织释放多种细胞因子和炎症介质,它们可加速颈椎间盘退变;而软骨终板钙化是颈椎间盘退变的始动因素,二者呈高度正相关。抗骨增生胶囊与葡立胶囊联合作用治疗退变颈椎间盘对IL-1β、TNFα含量及软骨终板钙化有明显的抑制作用。Objective:To explore the changes of cytokines in degenerated cervical intervertebral discs and the relationship between cartilage end-plate calcification of cervical spine and the degeneration of cervical intervertebral discs,and also to study the effect of combination treatment with Chinese and western medicine on cytokines and cartilage end-plate calcification.Methods:Twenty-five four-month-old New Zealand rabbits were made into dynamic imbalance model of cervical vertebra in order to induce cervical intervertebral disc degeneration. After 7 months, rabbits in treatment group were given anti-hyper osteogeny capsule and glucosamine hydrochloride capsule, twice a day for one month. Thereafter, all rabbits were sacrificed and the cervical intervertebral discs were collected. The contents of IL-1β and TNFα in cervical intervertebral discs in every group were determined by ELISA method, meanwhile, the degeneration of the cervical vertebrae and the thickness of the calcified layer of the cartilage end plates were also measured. Results;The contents of IL-1β and TNFα in degenerated cervical intervertebral discs and the thickness of the calcified layer of the cartilage end-plate in anti-hyperosteogeny capsule combined with glucosamine hydrochloride capsule group declined significantly compared with model group (P〈0. 05). Conclusion : Anti-hyperosteogeny capsule in combination with glucosamine hydrochloride capsule can decrease the contents of IL-1β and TNFα in degenerated cervical intervertebral discs and the thickness of the calcified layer of the cartilage end-plate in eervical spine.
