P27^(kip-1)蛋白对支气管哮喘大鼠模型气道平滑肌细胞凋亡的影响

摘　　要：目的探讨P27^(kip-1)对哮喘大鼠模型气道平滑肌细胞凋亡的影响。方法清洁级(SPF)雄性SD大鼠30只,随机分成对照组、哮喘4周组、哮喘6周组,每组10只。用卵清蛋白(OVA)致敏与激发建立大鼠哮喘模型,HE染色观察和测量气道形态学变化;末端转移酶介导的dUTP切口末端标记法(TUNEL)检测ASMCs凋亡并计算凋亡指数(AI);免疫组化法检测P27^(kip-1)蛋白表达。结果哮喘4周和6周组支气管壁厚度、支气管平滑肌厚度均显著高于对照组(P<0.01),且6周组高于4周组(P<0.01);哮喘4周和6周组凋亡指数均显著低于对照组(P<0.01),且6周组低于4周组(P<0.01);哮喘4周和6周组P27^(kip-1)蛋白表达显著低于对照组(P<0.05)。结论哮喘大鼠模型气道平滑肌细胞凋亡减少,可能与P27^(kip-1)蛋白表达减少有关。Objective To investigate the effects of P27^kip-1 on apoptosis of airway smooth muscle cells in asthmatic rats. Methodes 30 healthy,male SD rats were randomly and equally divided into three groups：control group, asthmatic 4 weeks group, asthmatic 6 weeks group. A rat asthmatic model was exploited, in which rats were sensitized with ovalbumin and Al（OH）3, and repeatedly exposed to aerosolized ovalbumin. After the last challenge, the animals were killed,The airway wall thickness （Wat/Pbm） and the bronchial smooth muscle thickness （Wam/Phm） were measured by image analysis system. The apoptosis in ASMCs was deteced by the technique of TdT- mediated dUTP- biotin nick end labeling （TUNEL）. The expression of P27^kip-1 was determined by immuneohistochemical staining. Results （1）The airway wall thickness,bronchial smooth muscle thickness, the airway resistance in two asthmatic groups were significantly higher than those in control group, respectively （P 〈 0.01 ） , and these indicators in asthmatic 6 weeks group was higher than those in asthmatic 4 weeks group, respectively （ P 〈 0.01 ）. （2） The apoptosis index （AI） of ASMCs in two asthmatic groups was decreased, as compared with group C,whose AI showed nomal （P 〈0.01 ）. （3）The expression of P27^kip-1 protein in two asthmatic groups was significantly lower than that incontrol group （ P 〈 0. 05 ） , and the indicator in asthmatic 6 weeks group was lower than that in asthmatic 4 weeks group （ P 〈 0.05 ）. Conclusion The protein P27^kip-1 possibly participaties in apoptosis of airway smooth muscle cells.
