苏云金芽胞杆菌杀虫晶体蛋白基因cry1E的改造

摘　　要：利用ＰＣＲ片段延伸连接技术将ｃｒｙ１Ｅ的原启动子准确地更换为ｃｒｙ３Ａ启动子，重组ｃｒｙ１Ｅ经载体ｐＨＴ３１５克隆至枯草芽胞杆菌无芽胞突变株１Ｓ８和苏云金杆菌以色列亚种无晶体突变株４Ｑ７，获得重组菌３Ａ１ＥＢｓ和３Ａ１ＥＢｔ。聚丙稀酰胺凝胶电泳和生物测定表明，克隆改造后的ｃｒｙ１Ｅ可表达，表达的Ｃｒｙ１Ｅ对海灰翅夜蛾有毒（幼虫死亡率６０％～７０％）。The original promoter of cry 1E was substituted with cry 3A promoter by PCR segment overlaping extension technique. The recomlinant cry 1E was inserted to pHT315 and transformed into Bacillus subtilis mutant 1S 8 and Bacillus thuringiensis subsp isrealensis acrystalliferous mutant 4Q7. The recombinant strain 3A1EBs and 3A1EBt were obtained. SDS PAGE and bioassay showed that modified cry 1E was toxic to Spodoptera littoralis (mortality of larva was 60%～70%).
