摘  要:目的:探讨细胞代谢相关基因在子痫前期胎盘组织中的表达谱及其影响机制。方法:使用含12000个与代谢、凋亡、细胞黏附、信号传导、转录因子等有关基因的cDNA表达谱芯片,检测4例妊娠期高血压疾病子痫前期及正常的胎盘组织的基因表达谱差异,并对部分差异表达基因进行了Northern验证。结果:4例子痫前期胎盘中共有44条基因表达发生了变化,其中糖原磷酸化酶(GP-M)基因、瘦素(leptin)基因、脂蛋白酯酶(LPL)基因及糖代谢相关基因(glucose transporter)表达增强,核苷酸代谢基因(CD73)与能量代谢调节基因(creatine kinase B)表达下降。结论:多种基因表达异常与子痫前期的病理发生有关,细胞代谢相关基因表达异常可能是血管内皮细胞损伤的原因之一。Objective:To study the expression of cyto-metabolism related genes in preeclamptic placenta and the possible mechanism involved in the regulation process. Methods: 12000 clones from the IMAGE library were used to build the cDNA microarray, which are involved in the functions of signal transduction, cell cycle, transcription, neuroprotection, apoptosis, angiogenesis, metabolism, extracellular matrix, and others. Complementary DNA (cDNA) probes were generated from human placentas biopsies of normal and preeclamptic pregnancies. After co-hybridized with the cDNA microarray, slides were scanned and the results were analyzed by software. To validate the cDNA microarray results, we also performed Northern-blot analysis on a small subset of affected genes. Results: 44 genes' expressions were significantly difference between normal and preeclamptic placentas. Among of them, Glycogen phosphorylase gene, leptin gene, Upoprotein lipase gene, and Glucose transporter gene were up-regulated, while CD73 gene and Creatine kJnase B gene were down-regulated. Conclusions:The change of cyto-metabolism related genes expression in placenta might be involved in the pathogenesis of preeclampsia.
