摘 要:目的探讨大鼠脑创伤后海马区C—fos基因表达及对学习记忆功能的影响。方法建立Marmarou大鼠脑创伤模型,采用分子原位杂交技术和Morris水迷宫分别观察大鼠伤后海马区C—fos基因表达变化及空间学习记忆能力的改变。结果伤后30min海马区神经元即出现C—fosmRNA的表达,1~3h达高峰,伤后24h表达基本消失;Morris水迷宫实验,大鼠伤后存在空间学习记忆功能障碍。结论脑创伤可引起C—fos基因在海马区的表达上调,通过介导延迟性神经元细胞死亡,影响细胞间的信息传递,参与认知功能的损害。Objective To explore the effect of c-fos mRNA expression in the hippocampus on the impairment of learning and memory of rats following traumatic brain injury. Methods The model of traumatic brain injury was established in rats by using Marmarou' s method. Then, c-fos mRNA expression was detected by means of in situ hybridization and cognitive dysfunction evaluated by Morris water maze. Results After traumatic brain injury, c-fos mRNA expression was detected in the hippocampus at 30 minutes, reached peak at 1-3 hours and almost disappeared at 24 hours. Morris water maze test showed significant impairment of spatial learning and memory in rats. Conclusions Traumatic brain injury can induce up-regulated expression of c-fos mRNA in the hippocampus of the rats and lead to cognitive impairment by mediating delayed neural cell death and influencing intercellular signal transmission.
