摘 要:目的探讨高压氧(hyperbaric oxygen,HBO)在新生鼠缺氧缺血性脑损伤(hypoxic-ischemic brain damage,HIBD)神经修复中的作用机制。方法新生7日龄SD乳鼠30只,随机分为HIBD+HBO干预组、HIBD组和正常对照组,每组10只。干预组于建模后予以HBO治疗,连续7d。采用免疫组化技术检测各组骨形态发生蛋白4(BMP-4)及神经巢蛋白(nestin)在海马的表达;采用原位杂交技术检测各组BMP-4及nestin mRNA在海马的表达;采用TUNEL方法检测各组神经细胞凋亡。结果3组中,干预组BMP-4蛋白在海马的表达最多,HIBD组BMP-4蛋白在海马的表达最少;干预组nestin蛋白在海马的表达最多,HIBD组nestin蛋白在海马的表达最少;干预组BMP-4mRNA在海马的表达最多,HIBD组BMP-4mRNA在海马的表达最少;干预组nestin mRNA在海马的表达最多,HIBD组nestin mRNA在海马的表达最少;HIBD组凋亡神经细胞最多,HBO干预组凋亡神经细胞与HIBD组差异有统计学意义(P<0.01)。结论HBO对HIBD具有神经修复作用,其作用机制可能与促进BMP-4及nestin在海马的表达以及抑制神经细胞的凋亡有关。Objective To investigate the function mechanism of hyperbaric oxygen(HBO) in neural repairation of neonatal rats after hypoxic-ischemic brain damage(HIBD).Methods Thirty 7-day-old rats were randomly divided into control group,HIBD group and HIBD+HBO group,with 10 rats in each.Bone morphogenetic protein-4(BMP-4)and nestin protein in the hippocampus were determined with immunohistochemical method while their messenger ribonucleic acid(mRNA) were determined with in situ hybridization.Apoptosis of nerve cells was determined with TUNEL. Results Expression of BMP-4 and nestin protein in the hippocam- pus was the highest in HIBD + HBO group while their expression in HIBD group was lower than that in control group. Expression of BMP-4 and nestin mRNA in the hippocampus was the highest in HIBD + HBO group while their expression in HIBD group was lower than that in control group. Apoptosis of neural cells in HIBD group was the highest among the 3 groups while apoptosis of neural cells in HIBD + HBO group was lower than that in HIBD group(P 〈0. 01). Conclusion HBO plays a role in nerve reparation, probably by promoting expression of BMP-4 and nestin in the hippocampus and inhibiting apoptosis of neural cells.
