摘 要:目的:运用蛋白质组学方法比较正常与糖尿病性大鼠心肌缺血再灌注损伤时心肌蛋白质组表达图谱的差异。方法:将SD大鼠随机分为心肌缺血再灌注组(IR组)和糖尿病心肌缺血再灌注组(DMIR组)。分别取各组左心室心肌蛋白进行二维电泳及凝胶染色后,用ImageMaster 2D Platinum软件对获得的蛋白图谱加以分析,寻找差异蛋白点。结果:二维电泳结果显示,IR组心肌蛋白二维电泳图谱与DMIR组相比共有29个显著差异点,其中6个点表达上调,6个表达下调,17个表达不匹配。结论:IR组与DMIR组大鼠心肌的二维电泳结果显示蛋白表达图谱有明显差异,但其差异蛋白为何种蛋白、起何种功能需要进一步的质谱数据进行分析。Objective: To analyze the differential expression of myocardial proteins of diabetic and non-diabetic rats with myocardial ischemia-reperfusion injury. Methods: Rats were randomly assigned to myocardial ischemia-reperfusion group (IR group) and diabetic myocardial ischemia-reperfusion group(DMIR group). The left ventricular myocardial protein was extracted and two dimensional polyacrylamide gel electrophoresis (2-DE) was followed with ImageMaster 2D Platinum software analysis in order to find the differences of the protein spots. Results: The result of two-dimensional electrophoresis showed that there were 29 protein spots expressed differently, of which 6 spots showed high expression, 6 spots showed low expression and 17 spots only expressed in IR group. Conclusion: There are significant differences between the myocardial protein expres-sion of IR group and DMIR group, but what’s the differential proteins and which role they played is still needed to further analyse by mass spectrometry.
