摘  要:目的利用量子点免疫荧光标记技术检测窖蛋白-1(Cav-1)在人肺癌组织芯片中的表达,探讨Car—1与细胞外基质金属蛋白酶诱导物(CD147/EMMPRIN)及基质金属蛋白酶-2(MMP-2)表达的关系。方法利用量子点免疫荧光组织化学结合组织芯片技术检测Cav-1、CD147和MMP-2在70例肺癌和5例非癌变肺组织中的表达。结果肺癌组Cav-1平均荧光强度为55±23,低于对照组的80±4(t=2.461,P=0.016);Cav-1表达与肺癌患者的性别、年龄和组织学类型无关,但与TNM分期(t=2.466,P=0.016)和淋巴结转移(t=2.972,P=0.004)均显著相关;Cav-1与CD147表达呈显著负相关(r=-0.331,P=0.005),与MMP-2表达无关。结论量子点免疫荧光组织化学技术可精确定量检测肺癌组织芯片上的不同蛋白表达。Car-1与肺癌的发生密切相关,其高表达可促进肺癌细胞的侵袭转移,其机制可能与调节CD147而不是MMP-2的活性有关。Objective Fluorescent semiconductor nanocrystals [ also known as quantum dots (QDs) ] are nanometer-sized light-emitting particles and are emerging as a new class of fluorescent probes for cancer detection, due to their unique optical and electronic properties. The aim of this study was to investigate the expression of eaveolin-1 ( Cav-1 ) , extraeellular matrix metalloproteinase inducer ( CD147/ EMMPRIN), matrix metalloproteinase-2 (MMP-2) proteins in the human lung cancer tissue microarray (TMA) by QDs immunofluoreseenee histoehemistry (QDs-IHC) and therefore to evaluate the relationship between Cav-1 protein and lung cancer invasiveness and metastasis. Methods QDs-IHC combined with TMA were used to detect the expression of Cav-1, CD147 and MMP-2 proteins in 70 cases of human lung cancers and 5 cases of noncancerous lung tissues. Results The average immunofluorescence intensity of Cav-1 protein in the lung cancer group was 55 ± 23, significantly lower than that in the control group (80 ± 4, t =2. 461, P =0. 016). The expression of Car-1 was not associated with the age and the gender of the patients, nor with the histology type of lung cancer ( P 〉 0. 05 ). The average immunofluorescence intensity of Cav-1 protein was associated significantly with TNM staging (t = 2. 466, P = 0. 016) and lymph node metastasis (t = 2. 972, P = 0. 004). A negative correlation was observed between Car-1 and CD147 protein expression(r = -0. 331 ,P = 0. 005 ), but no correlation was observed between Cav-1 and MMP-2 protein expression (P = 0. 193 ). Conclusions QDs-IHC could accurately and quantitatively detect different protein location in lung cancer TMA. A close relationship was detected between Cav-1 protein and the development of lung cancer. High expression of Cav-1 may be involved in invasiveuess and metastasis of lung cancer, possibly through the regulation of CD147 rather than MMP-2 activition.
