摘  要:目的观察异丙酚对缺血-再灌注心肌白介素-8(IL-8)和核转录因子-κB(NF-κB)的影响,探讨异丙酚减轻心肌缺血-再灌注损伤的机制。方法雄性新西兰大耳白兔24只,随机分为假手术组(Sham组)、缺血-再灌注组(I-R组)和异丙酚组(P组),每组8只。实验结束取缺血区心肌组织,免疫组织化学方法检测NF-κB、IL-8的表达;流式细胞术检测心肌细胞凋亡。结果I-R组、P组NF-κB、IL-8表达及凋亡细胞数均高于Sham组(P<0.05),P组各指标较I-R组降低(P<0.05)。结论抑制缺血-再灌注心肌NF-κB激活,进而降低IL-8表达,是异丙酚减少缺血-再灌注心肌细胞凋亡的机制之一。Objective To investigate the effects of propofol on the expression of nuclear factor kappa B (NF-KB) and the concentration of interleukin-8(IL-8) in arterial plasma by myocardial ischem I a-reperfusion injury in rabbits. Methods 24 New Zealand white rabbits weighted 2.1 - 2.3kg were divided into three groups randomly( n = 8 in each group) :group I (sham operation) ;group Ⅱ(I-R) and group Ⅲ(propofol + I-R). After the experiment the immunohistochemistry was used to detect the expression of NF-κB and IL-8 and flow cytometry (FCM)was used to detect the percentage of apoptotic cardiocytes. Results The expression of NF-κB,IL-8 and the percentage of apoptotic cadiocytes in group Ⅱ and group Ⅲ were significantly higher than those of group I(P〈0.05), and which in group Ⅲ were significantly lower than those in group Ⅱ(P〈0.05). Conclusion Propofol can inhibit the apoptosis of myocardium induced by ischemia - reperfusion injury by inhibiting the activation of NF-κB and down-regulating the levels of IL-8 ,which may be one of the action mechanisms of the effect of propofol.
