摘  要:目的该文探索建立了小鼠离体子宫收缩模型,以此为对象,对药物效应定量方法进行了评价研究,并与大鼠模型进行了比较。方法缩宫素诱导离体子宫收缩,观察10-8.5~10-5mol·L-1浓度范围内维拉帕米对子宫收缩的抑制作用,采用两次归一化的方法对数据进行处理,计算各浓度的抑制率,并根据实验结果计算药物的ED50,以上述方法观察维拉帕米对大鼠及小鼠离体子宫的不同作用。结果随着维拉帕米浓度增加,其对离体子宫收缩抑制作用不断增强,在10-5mol·L-1浓度下子宫收缩被完全抑制。两次归一化的方法对结果进行计算后,得到维拉帕米对大鼠及小鼠离体子宫收缩抑制作用的ED50分别为10-7.24(10-7.46~10-7.04)mol·L-1及10-7.58(10-7.78~10-7.38)mol·L-1。结论采用本方法进行实验及数据处理,可以较好的排除多种因素的干扰,从而对药物效应的强弱进行量化。经工具药物进行验证,认为该方法灵敏、可靠、经济,可以用于离体子宫活性药物的评价。Aim To establish an oxytocin induced in vitro uterine contraction model in mouse, to compare the model in rats, and to investigate the quantitative method of drug efficacy. Methods Oxytocin(5 U · L^-1 ) was used to induce the mouse uterine contraction. The contraction was recorded after treating with cumulative concentrations of verapamil ranging from 10^-8.5mol · L^-1 to 10^-5 mol· L^-1. The data of contraction were normalized twice by the data of control group;the effectiveness was reflected by the inhibitory rate. ED50 was then computed by this data. Effect of verapamil on rat uteri and mouse uteri was evaluated by the methods mentioned above. Results As the concentration of verapamil was raised , the inhibitory effect on mouse uteri increased. The contraction was suppressed completely once the verapamil concentration was raised to 10^-5 mol· L^-1. ED50 of verapamil for rat uteri and mouse uteri was 10^-7.24( 10^-7.46 - 10^-7.04)moL · L^-1 and 10^-7.55(10^-7.78-10^-7.38)moL · L^-1 ,respectively. Conclusions The method of processing data used in this study was efficacious to eliminate different kinds of interfer-ence. After verification, it was considered to be sensitive, reliable, economical, and useful to evaluate the effect of agents on uterine contraction in vitro.
