摘  要:【目的】观察针刺对抑郁模型大鼠海马脑源性神经营养因子(BDNF)和神经生长因子(NGF)表达的影响,探讨针刺的抗抑郁机制。【方法】选用SD大鼠随机分为正常组、模型组、针刺组、西药组,应用孤养和长期不可预见的慢性应激刺激复制抑郁大鼠模型,同时西药组按2 mg.kg-1.d-1剂量给予氟西汀灌胃,针刺组予以针刺百会、心俞、肝俞穴,每天1次,共21 d。采用免疫组化方法检测各组大鼠海马BDNF和NGF蛋白表达的情况。【结果】与正常组比较,模型组BDNF和NGF阳性细胞数量显著减少(P<0.01),而针刺组、西药组BDNF和NGF阳性细胞数显著增加(与模型组比较,P<0.05)。【结论】针刺可以增加抑郁大鼠海马BDNF与NGF表达,这可能是针刺抗抑郁的分子机制之一。Objective To observe the effect of acupuncture on brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF) expression in the hippocampus of depression rats, and to explore its possible anti-depression mechanism. Methods SD rats were randomized into the normal group, model group, acupuncture group and western medicine group. Rat models of depression were established by chronic unpredictable mild stress and by isolated raising. The western medicine group received gastric gavage of fluoxetine hydrochloride ( in the dose of 2 mg. kg^-1.d^-1), and the acupuncture group received acupuncture on acupoints of Baihui (GV20), Xinshu (BL15) and Ganshu ( BL17), once per day for 21 days. After treatment, the expression of BDNF and NGF in the hippocampus of rats was detected by immunohistochemical assay. Results The number of cells with BDNF and NGF positive expression in the hippocampus of model group was decreased (P 〈 0. 01 compared with the normal group), while was increased in the acupuncture group and western medicine group ( P 〈 0.05 compared with the model group). Conclusion The possible antidepressant mechanism of acupuncture is related with the increase of protein expression of BDNF and NGF in the hippocampus of depression rats.
