摘  要:目的优化小鼠前体成骨细胞体外分离培养方法,提高分离效率。方法用胶原酶A和分散酶消化出生72h以内的小鼠颅盖骨,分离获得的细胞通过光镜观察、碱性磷酸酶活性测定、Von Kossa染色和茜素红染色方法鉴定。结果获得的细胞活力高且具有典型的成骨特性。结论优化酶消化法是一种方便、高效分离小鼠前体成骨细胞的方法。[Objective ] To optimize the method to isolate primary osteoblasts from new born mice efficiently. [ Methods ] Osteoblasts were obtained cleanly by digestion of collagense A and dispase from 72h new born mice. Typical morphological and biological characteristics of the osteobiasts were identificated by observation under optic microscope, assay of Alkaline phosphatase (ALP) activity, ALP staining, Von Kossa staining and Alizarin Red staining. [Results] The results showed that the isolated cells had typical morphological and biological characteristics of the osteoblasts. [ Conclusion ] The modified enzymatic digestion is advantageous and efficient without trypsinase and shearing cranial bone.
