金葡菌对人巨噬细胞系U937细胞Fas/FasL表达的影响

摘　　要：目的探讨金黄色葡萄球菌(简称金葡菌)诱导人巨噬细胞系U937细胞凋亡时Fas/FasL蛋白的表达及意义。方法当细胞∶细菌为1∶20时分别培养0,15,30,60和90min,采用Western blotting检测Fas/FasL蛋白的表达情况,应用SPSS10.0软件包对结果进行统计学分析。结果Fas/FasL蛋白的表达随着金葡菌感染时间的延长而增高。结论金葡菌能够上调Fas/FasL蛋白的表达,从而可以诱导U937细胞的凋亡。[ Objective] The relationship between the induced apoptosis and expression of heat stress protein 90kd （HSP90） in human monocytic U937 cells by Staphylococcus aureus （S. aureus） was investigated. [ Methods ] U937 were used in all experiments, and they were infected with S. aureus when U937： S. aureus was 0, 1：5, 1：10, 1： 20, 1：50 and 1：100 respectively for 30 rain or for different lengths of time （0 min, 15 min, 30 min, 60 rain and 90 min respectively） when U937：S. aureus was 1：20. The apoptosis rates were examined by Annexin V FTTC/PI staining. Expression of HSP90 was detected by western blot. [Results] S. aureus could induce apoptosis of U937 in a dose- and time-dependent manner. The expression of HSP90 was upregulated in a time of infecting and dose of S. aureus dependent manner. [ Conclusions ] It is believed that U937 apoptosis induced by S. aureus may be related to the increase of HSP90.
