摘 要:目的观察转染NGF基因的Schwann细胞(SCs)在慢性神经根嵌压伤后神经修复中的作用。方法建立大鼠腰神经根慢性嵌压伤的动物模型,体外培养、纯化SCs并转染NGF基因,检测基因表达后种植于神经根嵌压伤处,术后2周处死动物切取神经根组织,行western blot、组织形态学及免疫组织化学检测。结果转染NGF的SCs可以在体内长期存活并形成新的髓鞘,较生理盐水对照组及单纯SCs组显著促进慢性神经根嵌压伤后的神经修复。结论转染NGF基因的SCs在慢性神经根嵌压伤后的神经修复中具有显著的促进作用,为慢性神经损伤的基因治疗提供了新的方法。Objective To observe the effect of Schwann cells(SCs) modified by NGF gene on the nerve repair after chronic nerve roots compressed injury.Methods The rat model of chronic compressed injury on nerve roots was established by insetting spinous process.SCs modified by NGF gene were transplanted into the site of injuried nerve roots after detecting the gene expression in vitro.At the second week after trasplantation,the tissues obtained at the injuried site was examined by Western blot,histology and immunohistochemistry.Results SCs modified by NGF gene could survive for long-time,form new myelin sheath.Conclusion SCs modified by NGF gene can promote nerve regeneration of chronic nerve roots compressed injury and provides a new method for the treatment on chronic nerve injury.
