双氢青蒿素对辐射小鼠血液系统的保护作用

摘　　要：目的研究双氢青蒿素(Dihydroartemisinin,DHA)对辐射损伤小鼠血液系统的保护作用。方法40只昆明小鼠,随机分为5组(DHA低、中、高剂量组、空白对照组和正常组),除正常组外,其余各组给予4.5Gy60Co-γ射线全身照射,以外周血白细胞、血小板计数观察辐射后小鼠血液系统的变化,并筛选出药物的合适剂量;另取24只小鼠,随机分为3组(用药组、对照组、正常组),以此前确定的最适剂量于照射前灌胃给药,予9.0Gy60Co-γ射线全身照射,照射后第9天处死小鼠,通过骨髓病理组织学改变进一步观察对小鼠造血系统的影响。结果双氢青蒿素使辐射后小鼠血小板升高(P<0.01),可降低死亡率,促进骨髓增生,对白细胞无明显影响。结论双氢青蒿素促进辐射小鼠血小板恢复,对小鼠血液系统有保护作用。Objeetive To study the protection effect of Dihydroartemisinin on bone marrow injury of mice indused by radiation damage. Methods 40 male mice were randomly divided into five groups （low,middle,high dose groups,control group and normol group ）,and were irradiated with 4.5Gy whole-body γ-ray except the normol group, detected the primary effeet on hematogical system by peripheral leukocyte and platelet count,and selected the most suitable dose of the drug.Then,twenty-four mice were randomly divided into three groups （drug treatment group,control group,normol group）, all were irradiated with 9.0Gy γ-ray except the normol group.Then killed all the mice 9 days after radiation, count spleen noduses,marrow nucleated cells,and detected morphology of marrow patho-tissue to observe the further effect on hematopoietie system. Results Dihydroartemisinin could promote the platelet on radiation damaged mice （P〈0.01）,decreased death rate （P〈0.01）,and showed no distinct influence on white blood cell. Conclusion Dihydroartemisinin can promote platelet to a normol level on radiation damaged mice, and protect hematological system.
