摘 要:目的了解在模拟HCV HVR1免疫7肽(7P)的刺激下,正常人免疫细胞分泌γ干扰素(IFN-γ)、白细胞介素10(IL-10)、肿瘤坏死因子α(TNF-α)及白细胞介素4(IL-4)的细胞频数变化,并探讨其作用机制。方法分离正常人外周血单核细胞(PB-MCs),分为CD4+CD8-T细胞组、CD4-CD8+T细胞组、NK细胞组、NKT细胞组四组,各组设阴性对照及豆蔻酸-佛波醇-乙酸酯(PMA)阳性对照,与7P共同孵育后,应用IL-10、IFN-γ、TNF-α、IL-4流式抗体进行细胞内因子检测,采用FACSCalibur流式细胞仪及FACS Calibur软件进行检测分析。结果与阴性对照组相比,分泌IFN-γ的细胞频数在NK细胞、NKT细胞以及CD4+CD8-、CD4-CD8+T淋巴细胞均明显增高(P<0.01);分泌IL-4的细胞频数变化不明显;分泌IL-10的细胞频数在CD4+CD8-、CD4-CD8+T淋巴细胞表现为升高(P<0.01,P<0.05),在NK细胞、NKT细胞表现为下降(P<0.01);分泌TNF-α的细胞频数在CD4+CD8-T淋巴细胞表现为下降(P<0.01),在NK细胞表现为升高(P<0.01)。结论7P能够通过调节细胞因子分泌改变免疫细胞的免疫反应方式,这种调节在以NK、NKT细胞为代表的固有免疫细胞和以CD4+CD8-、CD4-CD8+T细胞为代表的适应性免疫细胞之间存在差异,可能有利于在清除病毒的同时缓解炎症反应。Objective To demonstrate the frequency changes in immune cells on secreting interleukin-10 (IL-10),interferon-γ (IFN-γ),tumor necrosis factor-α (TNF-α) and interleukin-4 (IL-4) under the stimulation of a mimic 7-amino acid peptide of HCV HVR1 (7P),and to explore the mechanism of such changes. Methods The peripheral blood mononuclear cells (PBMCs) were isolated from healthy people and co-cultured with 7P. Intracellular cytokine staining was performed using antibodies against IL-10,IFN-γ,TNF-α and IL-4. The related cells were harvested on a FACSCalibur flow cytometer,and then analyzed with FACSCalibur software. Results Compared to the controls,the frequency of immune cells secreting IFN-γ was significantly increased in both CD4+CD8-,CD4-CD8+ T cells and NK,NKT cells (P0.01),while no change was found in the frequency of IL-4 secreting immune cells. The frequency of IL-10 secreting immune cells was significantly increased in CD4+CD8-and CD4-CD8+ T cells (P0.01,P0.05),while decreased in NK and NKT cells (P0.01). The frequency of TNF-α secreting cells was decreased in CD4+CD8-(P0.01),while increased in NK cells (P0.01). Conclusions The immune reaction model can be changed by 7P through modulating the secretion of cytokines in different immune cells,and there exists different modulation effects between the adaptive immunity cells,such as the CD4+CD8-,CD4-CD8+ T cells,and the innate immunity cells,such as the NK and NKT cells. Changes in cytokines secretion might contribute to the anti-virus action,and meanwhile lessen the inflammatory reaction.
