摘  要:目的建立快速简便的泛内酯、泛解酸的检测方法,方便酶法拆分DL-泛内酯过程中监测,研究pH对DL-泛内酯自发水解的影响。方法建立了用硅胶薄层色谱法以碘蒸汽为显色剂进行薄层色谱检测泛内酯和泛解酸;结合HPLC法建立了以旋光法评价酶法拆分DL-泛内酯反应程度的新方法;考察pH值对混旋泛内酯自发水解的影响。结果与结论薄层色谱和旋光测定的方法适合酶法拆分DL-泛内酯反应程度的快速检测;混旋泛内酯的自发水解随pH增加而增加,而且在一定pH值下,自发水解随着时间的延长而增加。Objective To establish quick and easy detection methods of pantolactone and pantoic acid, and study autohydrolysis status of DL-pantolactone during enzymatic resolution. Methods Pantolactone and pantoic acid were detected by using silica gel thin-layer chromatography with iodine vapor as a color developing agent;a new polarimetric method was established for the evaluation of enzymatic resolution reaction degree of DL-pantolactone by HPLC and the effect of pH in DL-pantolactone's autohydrolysis was studied. Results and Conclusions The methods of thin-layer chromatography and polarimetry are suitable for enzymatic resolution of DL-pantolactone reaction quick detection; DL-pantolactone's autohydrolysis increases with the increase of pH values; at a certain pH value, the autohydrolysis increases with the prolongation of the time duration.
