摘  要:目的探讨异氟烷对老年大鼠海马胆碱乙酰转移酶(choline acetyltransferase,ChAT)表达的作用和空间学习记忆的影响。方法20月龄老年雄性SD大鼠54只,随机分为3组(n=18)。A组(对照组):吸入30%氧气的空氧混合气体6h;B组(异氟烷单次吸入组):单次吸入1.2%异氟烷和30%氧气的空氧混合气体6h;C组(异氟烷多次吸入组):吸入1.2%异氟烷和30%氧气的空氧混合气体2h,1次/d,连续3d。各组处理中随机抽取5只行血气分析,5只完成处理24h后处死取脑分离海马,免疫组化法和RT-PCR法检测ChAT的表达,剩余行Morris水迷宫实验检测大鼠的空间学习记忆功能,持续6d。结果与A组比较,B、C组海马ChAT mRNA及蛋白表达下调(P<0.05);2~5d的逃避潜伏期延长(P<0.05);学习记忆曲线后移,B、C组至第4天渐趋稳定,而A组第3天趋于稳定;停留原平台象限的时间和穿越原平台次数减少(P<0.05);B、C组差异无统计学意义(P>0.05)。结论异氟烷可引起老年大鼠短期空间学习记忆功能减退,其机制可能与海马ChAT表达下调有关。Objective To explore the effect of isoflurane on the expression of choline acetyhransferase (CHAT) in the hippocampus and spatial learning and memory function in aged rats. Methods A total of 54 20-month Sprague-Dawley male rats, were randomly divided into 3 groups with 18 rats each group. Group A (control group) received a breath of 30% oxygen and air mixed gas for 6 h; Group B (Single inhalation of isoflurane group) were anesthetized with 1.2% isoflurane, 30% oxygen and air mixed gas for 6 h; Group C (Repeated inhalation of isoflurane group) were anesthetized with 1.2% isoflurane, 30% oxygen and air mixed gas 2 h every day for 3 d. Five rats were randomly selected from each group for the arterial blood gas analysis in the course of treatment. Another five rats randomly selected from each group were killed and their hippocampus were immediately isolated for detection of ChAT expression by immunohistochemical assay and RT-PCR in 24 h after treatment. The spatial learning and memory function of 24 remained rates was assessed with Morris watermaze test form 1 d to 6 d after treatment. Results The expression levels of mRNA and protein of ChAT in the hippocampus was significantly lower in group B and group C than group A ( P 〈 0.05 ). At day 2, 3, 4 and 5 after treatment, the escape latency in group B and group C was observed significantly longer compared with that of group A (P 〈 0. 05). Learning and memory curve was shift right in group B and group C, and turned to be stable at the 4th day but at the 3rd day in group A. The results of Morris water-maze test showed that the frequency of crossing the original platform and the time consumption of staying the original platform quadrant was lower in group B and group C than those in group A (P 〈 0.05 ). No significant differences was found between group B and group C ( P 〉 0.05 ).
