摘 要:[目的]观察丹参多酚酸盐对大鼠脑缺血再灌注损伤的影响。[方法]SD大鼠分为4组,分别为假手术组、缺血对照组、丹参多酚酸盐治疗组(10mg/kg)、丹参多酚酸盐治疗组(30mg/kg)。采用Longa法建立大鼠局灶脑缺血再灌注损伤模型(MCAO),正常对照组不做任何处理。丹参多酚酸盐治疗组在术后当天(术后60min)及术后d1,d2分别按设定剂量通过腹腔注射给予丹参多酚酸盐,丹参多酚酸盐溶于0.9%生理盐水中;假手术组和缺血对照组则腹腔注射生理盐水。各组大鼠在实验时根据实验设计在再灌注24h或72h处死,收集标本进行检测。TTC染色测定脑梗死体积,LONG法进行神经行为学评分,同时按照试剂盒说明检测血清中SOD,GSH-Px和MDA的变化。[结果](1)与缺血对照组比较,丹参多酚酸盐可以明显降低实验大鼠的行为学评分和减少的脑梗死体积,且丹参多酚酸盐(30mg/kg)组降低较丹参多酚酸盐(10mg/kg)组更为显著(P﹤0.05)。(2)与假手术组相比,缺血对照组、丹参多酚酸盐(10mg/kg)组和丹参多酚酸盐(30mg/kg)组血清中SOD和GSH-Px的活性明显下降(P﹤0.01),但丹参多酚酸盐能明显提升脑组织中SOD和GSH-Px的活性且随着药物剂量的增大,SOD和GSH-Px的活性增加越明显(P﹤0.01)。(3)与假手术组相比,缺血对照组、丹参多酚酸盐(10mg/kg)组和丹参多酚酸盐(30mg/kg)组MDA的含量明显升高(P﹤0.01),但丹参多酚酸盐则能明显降低脑组织中MDA的含量且随着药物剂量的增大,MDA减少越明显(P﹤0.01)。[结论](1)丹参多酚酸盐能减少脑缺血再灌注损伤大鼠脑梗死体积和降低神经行为学评分,表明丹参多酚酸盐对脑缺血具有保护作用。(2)丹参多酚酸盐能使大鼠血清中SOD和GSH-Px的活性增加,而MDA含量下降,说明丹参多酚酸盐能缓解脑缺血再灌注损伤后自由基产生和清除系统的失衡,从而发挥脑缺血的保护作用。[Objective]To detect the protective effect of salvianolte on the focal cerebral ischemia-reperfusion injury in rats and to reveal the mechanism. [Methods] 48 SD rats were divided into four group randomly, sham control group, ischemia control group, salvianolate (10 mg/kg) group and salvianolate (30 mg/kg) group, each group contain 6 rats. We used the Longa method to establish the MCAO model. The salvianolte at different dose were injected introperitoneally at the different time point according to the test design. The cerebral infarction volume by TTC stain and behavior scores by Longa's methods were also determined in order to evaluated the protective effect. Also we used assay kit to determine the activities of SOD and GSH-Px, content of MDA in each group. [Results] (1) Compared with ischemia control group, salvianolate could obviously reduce behavior scores and cerebral infarction volume, and the results were more obvious in salvianolate (30 mg/kg) group than in salvianolate (10 mg/kg) group. (2) The activities of SOD and GSH-Px of serum in the ischemia control group, salvianolate (10 mg/kg) group and salvianolate (30 mg/kg) group were much lower than those in normal control group. But the salvianotes can inhibit this decrease. (3) The content of MDA of brain tissue in the ischemia control group, salvianolate (10 mg/kg) group and salvianolate (30 mg/kg) group were much higher than those in normal control group. The salvianolate can inhibit this increase. [Conclusion] Salvianolate plays neuroprotective effect maybe through modulating the imbalance of oxidant and antioxidant inducing by the cerebral ischemia-reperfusion.
