STUDY ON EFFECT OF MANGANESE ON CELL APOPTOSIS AND THE ANTAGONISM OF ZINC IN MICE

摘  要:[目的]研究锌对锰诱发小鼠肝、脑细胞凋亡的拮抗作用。[方法]60只昆明种小鼠随机分为5组,即对照组、染锰组和锰加低锌组、锰加中锌组、锰加高锌组,每组12只。对照组小鼠腹腔注射生理盐水;染锰组小鼠腹腔注射氯化锰;加锌组小鼠注射氯化锰并在饮水中添加不同剂量的硫酸锌。4周后用末端脱氧核苷酸转移酶介导的缺口末端标记(TUNEL)和DNA琼脂糖凝胶电泳方法检测小鼠肝、脑细胞凋亡情况。[结果]染锰组及低、中剂量加锌组小鼠肝、脑组织出现"梯形"带改变,而高剂量加锌组小鼠肝、脑组织未出现"梯形"带改变,染锰组小鼠肝、脑细胞中TUNEL阳性细胞率明显高于对照组(P﹤0.01);补充锌后,随着添加锌剂量的增加,小鼠肝、脑细胞中TUNEL阳性细胞率明显下降,其中高剂量加锌组小鼠肝、脑细胞中TUNEL阳性细胞率低于染锰组,差异有统计学意义(P﹤0.01);高于对照组,但差异无统计学意义(P﹥0.01)。[结论]过量锰可诱发小鼠肝、脑细胞凋亡;较高剂量的锌对锰的毒性作用具有拮抗作用。[Objective]To research the influence of manganese and the antagonistic action of zinc on apoptosis in brain and liver cells of mice. [Methods] Kunming mice were randomly divided into 5 groups:Control group, manganese group, manganese plus low-dose zinc group, manganese plus id-dose zinc group, manganese plus high-dose zinc group. Mice were injected intraperitoneally by manganese chloride (MnCl2) and zinc was given through the drinking water. Four weeks later, the apoptosis of brain and liver cells in mice were detected with DNA agarose gel electrophoresis and TUNEL technique. [Results] The results showed that DNA ‘Ladder’ fragmentation of apoptotic hepatocytes were shown on agarose gel electrophoresis in the manganese group, manganese plus low-dose zinc group, manganese plus id-dose zinc group, and they weren not shown in manganese plus high-dose zinc; compared with the control group, the number of apoptotic cells in the brain and liver was obviously increased in manganese group (P﹤0.01). After adding zinc, the number of apoptotic cells was lower than that of the groups exposed to manganese. The number of apoptotic cells in manganese plus high-dose zinc group were significantly lower than that of manganese group (P﹤0.01). [Conclusion] Manganese can cause cell apoptosis in brain and liver cells. However, those effects can be antagonized by zinc.
