摘 要:以15-羟基十五烷酸和甲醇为主要原料,经酯化反应制备了15-羟基十五烷酸甲酯中间体,采用脂肪酶作为催化剂,15-羟基十五烷酸甲酯进一步经内酯化反应制备了大环麝香香料环十五内酯。实验分别考察了溶剂效应、酸碱效应、反应时间、水的体积分数、大孔树脂吸附提取等因素对内酯化转化率的影响,采用高效液相色谱法对产物进行检测。结果表明,以环己烷为反应溶剂,pH为6.5~7.5,反应时间控制在170 h,在微量水存在的条件下,结合大孔树脂吸附提取,环十五内酯的产率达到14.86%。In this paper, the cyclopentadecanolide was synthesized by an enzymatic esterification of the intermediate product of 15-hydroxy-15-alkylacid armourester, which was prepared with 15-hydroxy-15-alkyl acid and methanol. Using HPLC, the effect of solvent, acid-base effect, reaction time, volume fraction of water, and adsorption of macroporous resin on the conversion rate was determined. The result showed that the conversion rate could be up to 14.86% when the reaction was carried out for 170 h using cyclohexane as solvent at pH 6.5-7.5, followed an extraction on the macroporous resin with trace quantity water.
