摘 要:目的:研究灯盏花乙素苷元对大鼠实验性脑缺血的保护作用。方法:灯盏花乙素苷元(100,50,25 mg.kg-1)给大鼠连续灌胃7 d,采用电凝法阻断大鼠大脑中动脉(MCAO)制备局部脑缺血模型,缺血24 h后测定神经行为学评分和脑梗死面积;采用结扎双侧颈总动脉致全脑缺血模型,观测脑组织病理改变、脑含水量和脑组织中Na+,K+含量;实验同时设灯盏花乙素(100 mg.kg-1)阳性对照。结果:灯盏花乙素苷元能够明显改善MCAO局部脑缺血大鼠的神经功能障碍,缩小脑梗死面积;降低全脑缺血大鼠的脑含水量,抑制缺血脑组织的Na+潴留及K+流失现象,并可改善光镜下脑组织缺血性损伤。结论:灯盏花乙素苷元对大鼠实验性脑缺血模型具有保护作用,疗效优于灯盏花乙素。Objective: To investigate the protective effect of scutellarein on rat cerebral ischemia. Methods: After oral administration of scutellarcin 100, 50 or 25 mg·kg^-1 for 7 days, focal cerebral ischemia was induced in male Wistar rats by middle cerebral artery occlusion with the method of electric coagulation, and 24 hours later, the neurological deficit score and the cerebral infarct volume were assessed. Besides, in rats undergone same medication schema, global cerebral ischemia was induced by ligation of bilateral carotid and the pathological changes were assessed meanwhile the water content and Na^+ , K^+ concentration in brain were also measured. In both experiments, 100 mg·kg^1 scutellarin was tested as positive control. Results: Scutellarein ameliorated the neurological behavior and restricted the infarction development in MCAO rats in a dose-dependent manner. Also, it reversed the brain edema, the ionic disorder and the pathological changes in global cerebral ischemia. Conclusion: Scutellarein has protective effect, better than scutellarin, in rat cerebral ischemia.
