摘  要:目的:应用Aβ1-42孵育PC12细胞株制作细胞损伤模型,探讨α-硫辛酸体外抗β-淀粉样蛋白(Aβ)损伤的作用。方法:用Aβ1-42和α-硫辛酸对PC12细胞进行处理,MTT法检测细胞活性,测定细胞培养上清液中LDH的活性检测细胞膜的损伤,测定细胞Caspase-3活性检测各组细胞的凋亡。结果:经Aβ1-42处理的PC12细胞,活性降低。用α-硫辛酸保护处理可提高细胞的存活率,降低LDH释放及Caspase-3活性,具有显著性差异(P<0.01)。结论:α-硫辛酸可拮抗Aβ1-42诱导的细胞损伤,对神经细胞具有一定的保护作用。Objective: To investigate the protective effects of lipoic acid on the damage induced by amyloid-β peptidel 42 (Aβ1-42) in PC12 cells. Methods: PC12 cells were treated with Aβ1-42 and lipoic acid. The viability of PC12 cells was detected by MTT assay. The loss of membrane integrity and the cell lysis were measured by assaying the release of lactic dehydrogenase (LDH). The activity of caspase-3 in cells was measured by means of the cleavage of fluorogenic substrate Ac-DEVD-MCA. Results: Aβ142 induced apoptosis of PC12 cells and decreased cell viability. Lipoic acid improved the viability of PC12 cells, decreased LDH level in the culture media, and reduced the increase in caspase-3 activity. Conclusion: Lipoic acid can inhibit the cellular toxicity of Aβ; it may protect nerve cells.
