HPLC法和UV法测定阿德福韦酯片的含量比较

摘　　要：目的:比较HPLC法和UV法两种方法测定阿德福韦酯片的含量。方法:HPLC法:色谱柱为岛津C18色谱柱(规格150 mm×4.6 mm,粒度5μm),以乙腈-0.02 mol/L磷酸氢二钾溶液(用磷酸调节pH值为5.0)(40∶60)为流动相,检测波长为261 nm;UV法:以0.1mol/L盐酸溶液为溶剂,在258 nm波长处测定。结果:HPLC法:阿德福韦酯进样量在0.41～4.09μg范围内线性关系良好,r=0.9999;UV法:阿德福韦酯浓度在4.21～42.14μg/mL范围内线性关系良好,r=0.9999。结论:两种方法均可作为阿德福韦酯片的含量测定方法。Objective ：To compare the HPLC method and the UV mehtod for determining the content of Adefovir Dipivoxil Tablets. Methods：The HPLC method was performed on a C18 column （150 mm×4.6 mm, 5 μm）, with the wavelength of 261 nm and acetonitrile- 0.02 mol/L potassium phosphate dibasic solution （pH 5.0 adjusted with phosphate caid） as the mobile phase. UV method：to determine adefovir dipivoxil at the wavelengh of 258 nm in 0.1 mol/L hydrochloric acid solution. Results：HPLC：within the range of 0.41.-4.09 μg, there was a good linear correlation, r=-0.999 9. UV：within the range of 4.21-42.14μg/mL, there was a good linear correlation, r=0.999 9. Conclusion：Both two methods can be used to assay Adefovir Dipivoxil Tablets.
