摘 要:研究旨在对猪T细胞诱导型刺激物(ICOS)基因的cDNA序列进行克隆与分析。根据已报道的人ICOS基因cDNA序列设计引物,首次从猪脾脏组织总RNA中扩增出ICOS基因编码区全长cDNA序列,克隆于pMD18-T载体后进行测序并进行序列拼接,运用生物信息学分析DNA序列。结果表明:该基因编码区全长630bp,编码210个氨基酸,包含5个外显子。该序列与人全基因核苷酸序列及推导的氨基酸序列的同源性分别为80%和85%;与狗和小鼠的推导氨基酸序列的同源性分别为81%和75%。这为进一步研究该基因的结构特点和功能奠定了良好基础。To clone and analyze the porcine cDNA sequcence of Inducible T-cell co-stimulator (ICOS) gene, a pair of primers specific to porcine ICOS gene were designed and synthesized according to the reported human ICOS gene sequence and was used to amplify pig ICOS cDNA from total RNA of porcine spleen cells. Cloned into pGEM18 T vector, the PCR product was sequenced and analyzed by bioinformatics subsequently. Sequence analysis showed that the open reading frame of the porcine ICOS was 630 bp in length, and encoded a protein of 210 amino acids. Compared with human ICOS eDNA, the homology of nucleotide acid and amino acid were 80% and 85%, respectively; Compared with dog and mice, the homology of amino acid were 81% and 75%, respectively. This study was the first report of porcine ICOS complete gene in China, it will lay a sound foundation for further study on the structure and function of porcine ICOS.
