免疫毒素的制备及特异性淋巴细胞无反应性的诱导

摘　　要：为减损排斥反应性淋巴细胞克隆，采用异型双功能试剂ＳＰＤＰ、２ＩＴ连接ａｎｔｉＣＤ４、ＣＤ８单克隆抗体与天花粉毒素（ＴＣＳ），应用ＳｅｐｈａｃｒｙｌＳ２００分离游离ＴＣＳ，采用ＳＤＳＰＡＧＥ电泳及细胞毒性分析测定合成的免疫毒素的纯度和生物学活性。电泳结果显示，连接混合物中含有连接的免疫毒素、游离的单克隆抗体和ＴＣＳ，ＳｅｐｈａｃｒｙｌＳ２００可除去游离的ＴＣＳ。体外，免疫毒素可特异性杀伤大鼠脾细胞，其作用具有剂量依赖性。由此提示抗ＣＤ４。To diminish the specific lymphocytes that responsive to the rejection of allograft. Anti rat CD 4,CD 8 monoclonal antibodies and trichosnthin (TCS) was conjugated to immunotoxin by heterobifunctional reagent SPDP, 2 IT. The free TCS was removed from conjugates mixture by a column of Sephacryl S 200. The SDS PAGE and cytotoxic assay was used to measure the biological activity of immunotoxin. SDS PAGE showed the immunotoxin, free McAb and TCS were in the mixture of conjugation, and the free TCS can be separated by Sephacryl S 200. In Vitro, the lymphocytes of rat can be killed by antiCD 4,antiCD 8 immunotoxin. The kill capability was relay to the amount of immunotoxin. The authors consider that the immunology unresponsiveness can be induced by antiCD 4,antiCD 8 immunotoxin. That was useful in induced transplantation tolerance.
