摘 要:培养基成份是植物组织培养中愈伤诱导和植株再生的重要影响因子。除氮、磷、钾等大量元素和外源激素外,金属离子对植物组织培养的影响也很明显。本文以棉花MSB为基本培养基,分别加入不同浓度的Zn2+、Ni2+、Co2+、Ag+4种金属离子,研究它们对棉花愈伤组织的诱导和分化成苗的影响。研究表明,适当浓度的Zn2+、Ni2+和Ag+能减少最早出愈天数,提高愈伤诱导率和植株再生率,而Co2+加入的作用与此相反。Zn2+和Ni2+的加入还能抑制外植体的褐化,但效果不及Co2+。且5μmol.L-1的Zn2+和Ni2+在愈伤的诱导和分化成苗方面具有正向协调作用,是最佳组合。Medium nutrition is the important factor for callus induction and seedling differentiation in plant tissue culture.Metal ions are also essential for plant tissue culture besides macroelements and phytohormones.In this paper,effects of Zn2+,Ni2+,Co2+and Ag+on the callus induction and seedling differentiation of cotton were studied by adding them in basic medium MSB.The results indicated that Zn2+,Ni2+and Ag+shortened the time of callus induction and increased efficiencies of callus induction and plant regeneration. However,the role of Co2+was opposite to that in cotton tissue culture. Zn2+and Ni2+inhibited explants brown,which inhibition efficiency was lower than that of Co2+. In addition,when 5μmol·L^-1 Zn2+ was used together with 5μmol·L^-1 Ni2+,the effects of callus induction and seedling differentiation were the best.
