摘 要:对慢性缺氧3wk大鼠静脉注射钾通道开放剂克罗卡林(Cromakalim,150μg/kg)和钾通道阻断剂格列本脲(Glybenclamide,200μg/kg),以研究其对大鼠肺循环及心血管功能的影响。20只大鼠随机分为两组,均给予缺氧3wk。Ⅰ组先给予Glybenclamide,然后给予Cromakalim;Ⅱ组先给予Cromakalim,再给予Glybenclamide。结果:缺氧大鼠在先给予Glybenclamide后,其肺循环无明显变化,给予Cromakalim后可见肺动脉压明显降低;而先给予Cromakalim即可见缺氧大鼠肺动脉压明显降低,再给予Glybenclamide,则可逆转Cromakalim对肺血管的舒张反应,从而导致肺动脉压升高。这些结果提示,钾通道活性的改变在缺氧性肺动脉高压发病机制中起到十分重要的作用,Cromakalim可作为治疗肺动脉压的有效药物之一。Objective: To evaluate the effect of patassium channel opener Cromakalim and potassium channel blocker Glybenclamide on pulmonary circulation in rats model of chronic hypoxia for 3 weeks by intravenous bolus injection. Materials and Methods. Twenty wistar rats were divided into two groups at random. The rats in group I were first given Glybenclamide then Cromakalim and what was done in group II was contrary to that in group I . Results: Glybenclamide had no effect on the pulmonary circulation, whereas Cromakalim decreased the pulmonary arterial pressure (mPAP) and pulmonary vascular resistance (PVR) in rats of group I . Cromakalim had the same effect on pulmonary circulation in group II just as that had happened in group I , but Glybenclamide reversed the inhibitory effect of Cromakalim on hypoxic pulmonary hypertension in rats of groug II . Conclusion: The decrease of potassium channel activity plays an important role in the development of chronic hypoxic pulmonary hypertension. Cromakalim is one of the most effective drugs for the treatment of chronic hypoxic pulmonary hypertension.
