摘  要:目的探讨巨细胞病毒(CMV)感染与细胞凋亡的关系。方法利用凋亡细胞原位检测技术,结合PI染色法流式细胞DNA含量分析术,检测了体外CMVAD169毒株感染的宿主细胞(人胚肺成纤维细胞,HEL)及体内CMV性肝炎患儿肝组织内细胞的凋亡发生情况,并应用原位核酸分子杂交和图像分析技术观察了CMV感染对细胞凋亡调控基因bcl2和fas表达的影响。结果体内外CMV感染均能明显诱导细胞凋亡的发生,同时CMV感染使肝细胞及HEL细胞内的bcl2基因表达降低,而使fas基因表达上调。结论CMV感染诱导细胞凋亡可能是通过影响bcl2和fas基因的表达而实现的,这种凋亡规律的异常可能参与了CMV性疾病的发生与发展。Objective To explore the relationship between cytomegalovirus (CMV) infection and the frequency of apoptosis. Methods The authors detected the human embryonic lung (HEL) fibroblast cells infected with CMV in vitro and hepatic tissues of CMV hepatitis with in situ end labeling (TUNEL) apoptotic cell detection method and flow cytometric cell cycle analysis, and also investigated the expressions of bcl 2 and fas mRNA by means of in situ hybridization techniques and image analysis system. Results CMV infection enhanced apoptosis significantly in vitro and in vivo (both P <0.01). Meanwhile, higher expression of fas mRNA and lower expression of bcl 2 mRNA were found in hepatic tissues of CMV hepatitis patients and CMV infected HEL cells than that in the normal control. Conclusion These studies demonstrate that abnormal expressions of bcl 2 and fas mRNA may be involved in the pathological mechanisms of diseases assosiated with CMV infection.
