摘 要:目的探讨不同配伍组方加味桃核承气汤大鼠含药血清对胰岛素(Regular insulin,RI)诱导兔血管平滑肌细胞(Vascular smooth muscle cell,VSMC)的细胞周期及细胞凋亡的影响,寻找本方抗糖尿病动脉粥样硬化的最佳配伍组方。方法应用血清药理学方法,采用兔主动脉平滑肌细胞株,以RI诱导VSMC增殖,随机分为空白组、模型组、罗格列酮组、美迪康组、加味桃核承气汤组、活血组、泻热通下组、益气养阴组及活血-泻热通下组共9组,每组6孔,制备上述9组含药血清,采用碘化丙啶(Propidium Iodide,PI)单染色法,以流式细胞仪检测VSMC的凋亡率及细胞周期。结果各组在各细胞周期的DNA含量比较,总体差异显著(P<0.01);模型组、美迪康组细胞凋亡最少;罗格列酮组、活血组、活血+泻热通下组细胞凋亡较多;罗格列酮组、活血组、活血+泻热通下组促进凋亡作用(Sub-GO期)较加味桃核承气汤全方组显著(P<0.01)。结论促进糖尿病VSMC凋亡的药物有效成分,可能在加味桃核承气汤活血组、活血+泻热通下组当中含量更高。在促进糖尿病VSCM凋亡的机制方面,罗格列酮组可能与G0/G1、G2M期阻滞有关;活血+泻热通下组、活血组可能与G2M期阻滞有关。Objective To observe the effection of the rat serum with Modified Taohe Chenggi Decoction and its compo- sition on insulin-induced cell cycle and apoptosis of diabetic rabbit vascular smooth muscle cell. To discuss its possible mecha- nism of action and to search the best composition of Modified Taohe Chengqi Decoction in anti-atherosclerosis in diabetes melli- tus. Methods We used aorta smooth muscle cell line of rabbit. The vascular smooth muscle cell proliferation was induced by regular insulin. The different compositons of Modified Taohe Chengqi Decoction were made. We used Propidium Iodide single stain method and detected the cell cycle and apoptosis of diabetic vascular smooth muscle cell. Results Method of FCM showed that there was the least apoptosis in the model group and the Metformin group. And there was the most apoptosis in the TZDs group, the promoting blood circulation group and the promoting blood circulation and purging heat and stool group. As to the apoptosis promoting action, these three groups were significaritly different from the complete decoction group. Conclusion The active ingredients in apoptosis promoting of diabetic vascular smooth muscle cell might be more in the TZDs group, the promoting blood circulation group and the promoting blood circulation and purging heat and stool group. As to the mechanism of action, the TZDs group might be related to inhibition of G0/G1 phase and G2M phase. And the promoting blood circulation group and the promoting blood circulation and purging heat and stool group mighte be related to inhibittion of G2M phase.
