摘 要:探讨同型半胱氨酸(HCY)对大鼠主动脉内皮细胞(EC)分泌一氧化氮(NO)、血管紧张素Ⅱ(Ang Ⅱ)和内皮素(ET)的影响.取体重150—200g健康雄性Wistar大鼠的胸主动脉,以组织贴块法进行EC培养,传代至第5代用于实验,各实验组及对照组均为6孔细胞,实验组加入不同浓度的HCY,对照组不加HCY,分别在2、4、6、8、12、24h观察细胞形态并取细胞培养液测定NO含量.一氧化氮合成酶(NOS)活性、AngⅡ和ET含量.HCY可引起EC形态发生变化HCY刺激EC分泌NO、Ang Ⅱ和ET呈时间和剂量依赖性.提示:HCY使EC分泌NO、Ang Ⅱ、ET平衡失调可能是导致动脉粥样硬化的重要原因之一.Objective:To study the effects of homocysteine(HCY)on cultured rat aortic endothelial cells(ECs)secreting Nitric Oxide(NO) 、Angiotensin Ⅱ (Ang Ⅱ ) and Endothelin(ET) .Methods: Cultured rat aortic ECs were exposed to HCY at the concentrations of 0,0.5, l,2.5,5mM for 2,4,8,12 and 24h respectively, in the end of each experiments, the morphological changes were observed and the medium was collected to assay NOS activity and the concentration of NO, Ang Ⅱ , ET. Results :The morphological effects of exposing a confluent rat aortic ECs monolyer to 5mM HCY:ECs contracted gradually and detached from the substratum and few cells remain attached after 24h.HCY at the concentrations of 0、0.5、1、2、2.5mM were incubated with ECs for 24h,led to dose - dependent increase in NOS activity(P < 0.05)and NO(P < 0.05), Ang Ⅱ and ET did not increase significantly than the control group(P> 0.05) ,ECs were exposed to 5mM HCY for 2、4、6、8h,the secretion of NO inceased significantly(P < 0.05), Ang Ⅱ and ET did not increase significantly than the control group(P> 0.05) .Whereas, after 12h and 24h,5mM HCY resulted in impaired NO responses, the secretion of Ang Ⅱ and ET increased significantly than the control group(P< 0.05) .Conclusion: ECs secreting NO 、 Ang Ⅱ and ET imbalance may be one of the atherogenic mechanism of HCY.
