摘  要:目的观察附子对脊神经结扎大鼠痛阈的影响,并初步探讨其镇痛机制。方法24只雄性SD大鼠随机分为正常对照组、神经痛组和附子治疗组,每组各8只。左侧L_5脊神经结扎术后8 d,附子治疗组给予附子2 g/kg灌胃,连续5d,1次/d;神经痛组给予同等剂量的生理盐水。术前至术后12 d每天检测大鼠痛阈变化。术后12 d取各组大鼠左侧L_5和L_4及右侧L_5 DRG,观察DRG中交感神经纤维及篮状结构数量的变化。结果附子治疗组大鼠治疗后痛阈逐渐增高,与神经痛组比较差异具有统计学意义(P<0.05);附子治疗组大鼠左侧L_5和L_4 DRG中TH阳性纤维平均累计光密度明显低于神经痛组,右侧L_5 DRG中TH阳性纤维的平均累计光密度在附子治疗前后差异无统计学意义。附子治疗组DRG中篮状结构的数量较神经痛组减少,但两组比较差异无统计学意义。结论附子确能提高脊神经结扎大鼠的痛阈,其机制可能与抑制脊髓DRG中交感神经的芽生相关。Objective To study the effect of prepared aconite root on pain threshold of spinal nerve ligated rat model, and the analgesic mechanism of prepared aconite root. Methods Twenty - fourmale SD rats were randomly divided into three groups( n = 8 each) : normal group,spinal nerve ligated (SNL) group and prepared aconite root (PAR) group. In PAR group, PAR was administered orally once a day at a dose of 2 g/kg for 5 days from the eighth day after the left L5 spinal nerve was ligated. In SNL group, normal saline was administered in the same dose. The changes of pain threshold before and after surgery were observed once a day. At the 12^th day after surgery,the left L5 and L4 dersal root ganglia (DRG) as well as the right L5 DRG were removed to be immunostainied for observing the changes in numerical density of TH - IR fibers and "TH - IR - wrapped" neurons. Results The pain threshold of rats in the PAR group was significantly higher than that of the SNL group after PAR treatment. The extent of TH - IR fibers in the left L5 and L4 DRG of the PAR group were significantly lower than those of the SNL group,while there was no significant difference in the right L5 DRG after PAR treatment. The "TH - IR - wrapped" neurons in the PAR group were fewer in number than the SNL group, but there was no significantl difference between the two groups. Conclusions PAR improves the pain thresthold of the spinal nerve ligatied rats model. The inhibition of the growth of sympathetic sprouting in the DRG may be involved in the mechanism of relieving pain.
