摘 要:[目的]探索控制短叶巨象增殖继代次数的方法。[方法]以短叶巨象的单芽为外植体,以MS培养基为基本培养基对其进行离体培养,进而建立短叶巨象无性繁殖体系并在低温环境下长时间保存该离体种质。[结果]接种到诱导培养基上35~40 d后,外植体膨大且有愈伤组织形成;在培养基MS+Kt3.0 mg/L+NAA0.1 mg/L和MS+Kt2.0 mg/L+NAA0.1 mg/L中,从膨大的外植体外周长出了小芽。小芽接种到增殖培养基MS+Kt3.0 mg/L+NAA0.1 mg/L和MS+Kt2.0 mg/L+NAA0.1 mg/L中约30 d后又有新的丛生芽发生。小苗接种到生根培养基上20~30 d后,在培养基1/2 MS+NAA1 mg/L中生根较粗、较少;在培养基1/2 MS+IBA1 mg/L中生根较细、较多。冷藏4个月后的丛生芽的增殖苗及增殖效果与未经冷藏的种苗无明显差异。[结论]该研究为短叶巨象的快速商业化生产和种质保存奠定了技术基础。[ Objective ] The research aimed to explore the method of controlling the times of multiplication and successive transfer cuhure of Gasteria brevifolia. [ Method] With the simple buds of G. brevifolia as explants, they were cultured in vitro with MS culture media as basic culture media, so as to set up the asexual propagation system of G. brevifolia and these in vitro germplasm were conserved at low temperature for long time. [ Result] After 35 -40 d from the time when the explants were inoculated onto inducing culture media, they were expanded, there were calli formed on them and on the culture media MS + Kt 3.0 mg/L + NAA O. 1 mg/L and MS + Kt 2.0 mg/L + NAA 0.1 mg/L, there were gemmules grown out from the external of expanded explants. After about 30 d from the time when the gemmules were inoculated on- to the multiplication culture media MS + Kt 3.0 mg/L + NAA 0. 1 mg/L and MS + Kt 2.0 mg/L + NAA 0.1 mg/L, there were new clustered buds grown out. After 20 - 30 d from the time when the plantlets were inoculated onto rooting culture media, the roots grown out in the medium 1/2 MS + NAA 1 mg/L were thicker and less and that in the medium 1/2 MS + IBA 1 mg/L were thinner and more. The reproductive shoots and multiplication effect of clustered buds treated by cold storage for 4 months showed no obvious difference from that of seedlings non-treated by cold storage. [ Conclusion] This research laid a technological foundation for the rapid commercial production and germplasm conservation of G. brevifolia.
