摘 要:[目的]利用ISSR技术分析江苏地区5个三角帆蚌种群的遗传多样性和亲缘关系。[方法]用10条ISSR引物对5个三角帆蚌种群基因组DNA进行扩增,选取其中5条扩增条带多、信号强、背景清晰的引物对5个群体(无锡太湖、洪泽湖、南京江浦、兴化1、兴化2)共121个个体进行扩增分析。[结果]通过ISSR扩增共获得70个DNA片段,大小在200~2 000 bp,其中61个位点表现多态性,多态位点比例为87.14%。5个种群的基因多样性指数在0.259 9~0.314 4,平均值为0.290 1,Shannon信息指数在0.392 2~0.466 3。南京江浦养殖种群的2种遗传多样性指数最高分别为0.314 4和0.466 3,而无锡太湖野生种群的最低为0.259 9和0.392 2。[结论]三角帆蚌养殖种群的遗传多样性高于野生种群;5个三角帆蚌种群间的亲缘关系与地理位置无显著相关性。[ Objective ] The study aimed to analysis the genetic diversity and genetic relationship of 5 Hyriopsis cumingii populations in Jiangsu aera by using Inter-Simple Sequence Repeat (ISSR) technique. [ Method] The genome DNA of five H. cumingii populations were amplified by using 10 ISSR primers in which the five primers with more amplification bands, strong signal and clear background were selected to make the amplification analysis on 5 populations from Taihu in Wuxi City (TH) , Hongzehu ( HZH), Jiangpu in Nanjing (JP), Xinghua 1 ( XH1 ), Xinghua 2 (XH2) which was 121 individuals in total. [ Result ] The 70 DNA fragments were obtained and their sizes were from 200 to 2 000 bp through ISSR amplification, in which, the 61 sites were polymorphism and the proportion of polymorphic site was 87.14%. The gene diversity index of 5 populations were 0. 259 9 - 0.314 4 with the average of 0. 290 1 and the Shannon message index was 0. 392 2 - 0.466 3. The highest value for 2 kinds of the genetic diversity index in breed populations in JP were 0. 314 4 and 0.466 3 resp. , but the lowest value for that of wildness populations in TH were 0. 259 9 and 0. 392 2 resp. [ Conclusion ] The gene diversity of H. cumingii breed populations was higher than the wildness populations and the genetic relationship of 5 H. cumingii populations had no significant correlation with the geography position.
